| Abstract: | The respiratory syncytial virus (RSV) can cause serious cases of bronchiolitis and pneumonia, particularly in risk groups such as children under the age of five, immunocompromised and older people. Although there are now prophylactic drugs such as monoclonal antibodies and vaccines on the market, there is no way of treating acute infections. Therefore, it is necessary to develop small-molecule substances with anti-RSV activity, as their production is usually much faster and cheaper. The starting point of the present work was a total of ten smallmolecule classes, which were selected from the hits of a phenotypic high-throughput screening of approx. 42,000 compounds based on medicinal-chemistry considerations. These hits included seven entry inhibitors, one replication inhibitor and two inhibitors with an unknown mode of action. The present work focuses on their hit confirmation by purity analysis and resynthesis as well as their structure-guided optimization by design and synthesis of hit derivatives. With the exception of two compound classes, all hits were confirmed as genuine. These were investigated in more detail using purchased and synthesized derivatives. Due to interesting activity profiles, three classes were selected for more extensive studies and optimization work in the further course of this thesis, which led to several frontrunners and new starting points for future optimizations. Das Respiratorische Synzytial-Virus (RSV) kann bei Risikogruppen wie Kindern unter fünf Jahren, immunsupprimierten und älteren Menschen schwere Fälle von Bronchiolitis und Pneumonie hervorrufen. Mittlerweile gibt es zwar prophylaktische Arzneimittel wie monoklonale Antikörper und Impfstoffe auf dem Markt, allerdings dienen diese nicht der Behandlung akuter Infektionen. Daher ist es notwendig, insbesondere niedermolekulare Substanzen mit anti-RSV-Wirkung zu entwickeln, denn ihre Herstellung ist meist deutlich schneller und kostengünstiger. Ausgangspunkt dieser Arbeit waren insgesamt zehn niedermolekulare Substanzklassen, die aus den Hits eines phänotypischen Hochdurchsatzscreenings von rund 42.000 Verbindungen unter medizinalchemischen Aspekten ausgewählt wurden. Darunter befanden sich sieben Eintrittsinhibitoren, ein Replikationsinhibitor und zwei Inhibitoren mit unbekanntem Wirkmechanismus. Die vorliegende Arbeit fokussiert sich auf deren Hit-Bestätigung durch Reinheitsanalyse und Resynthese sowie deren strukturgeleitete Optimierung durch Design und Synthese von Hit- Derivaten. Bis auf zwei Verbindungsklassen wurden alle Hits als Echte bestätigt. Diese wurden mithilfe von eingekauften und synthetisierten Derivaten näher untersucht. Aufgrund interessanter Wirkprofile wurden drei Klassen im Verlauf dieser Dissertation für umfangreichere Studien und Optimierungsarbeiten ausgewählt, die neben mehreren Spitzenkandidaten zu neuen Startpunkten für zukünftige Optimierungen führten. |
