Abstract: | In dieser Arbeit wurde die zelluläre Ca2+-Kationenhomöostase in primären Sec61a1Y344H/Y344H-Hautfibroblasten aus Mus musculus durch Ca2+-bildgebende Verfahren analysiert. Auf der luminalen Seite des endoplasmatischen Retikulums kam ein reversibel Ca2+-schaltbares Fluoreszenzprotein, das GCaMP6-150-Konstrukt, zum Einsatz und im Cytosol der synthetisierte fluoreszierende, ratiometrische Ca2+-Indikator Fura-2. Besonderes Augenmerk galt dem passiven Ca2+-Flux vom Lumen des endoplasmatischen Retikulums ins Cytosol im Hinblick auf seine Kinetik. Unter basalen Bedingungen verhielten sich die Zellen mit Sec61a1Y344H-Fehlsinnmutation nicht anders als die Wildtypzellen, wurden diese jedoch über einen Zeitraum von 24 Stunden mit dem endoplasmatisches Retikulum-Streßor Tunicamycin behandelt, so wurde die zelluläre Ca2+-Kationenhomöostase nicht mehr gewahrt. Die Sec61a1Y344H/Y344H-Hautfibroblasten zeigten eine massive Zunahme des passiven Ca2+-Flux vom Lumen des endoplasmatischen Retikulums ins Cytosol bei gleichzeitig beschleunigter Kinetik. Da die Sec61a1Y344H-Fehlsinnmutation in Mus musculus unter anderem mit Diabetes mellitus Typ 1 assoziiert ist, eine gestörte Insulinbiosynthese und -sekretion jedoch nicht nachgewiesen wurde (Lloyd, Wheeler et al., 2010), wird auf Grund der in dieser Arbeit vorliegenden Daten eine mögliche Störung der zellulären Ca2+-Kationenhomöostase als alternativer/zusätzlicher pathobiophysikalischer Mechanismus des Sec61Y344H-assoziierten Diabetes mellitus Typ 1 in Mus musculus vorgeschlagen. In this doctoral thesis the cellular Ca2+ homeostasis in primary Sec61a1Y344H/Y344H-skin fibroblasts of Mus musculus was analysed by Ca2+ imaging. Two fluorescent Ca2+ indicators were used, the fluorescent protein GCaMP6-150 which was directed to the lumen of the endoplasmic reticulum and the synthetic ratiometric Ca2+ dye Fura-2 which was directed to the cytosol. Especially the passive Ca2+ flux directed from the lumen of the endoplasmic reticulum to the cytosol was analysed according to its kinetics. There was no difference between Sec61a1Y344H/Y344H-skin fibroblasts and wildtype cells under basal conditions. After treatment with tunicamycin for 24 hours the Ca2+ homeostasis was disturbed. The Sec61a1Y344H/Y344H-skin fibroblasts showed a massive rise in the passive Ca2+ flux with faster kinetics. In Mus musculus the Sec61a1Y344H-missense mutation leads to type 1 diabetes mellitus among other things (Lloyd, Wheeler et al., 2010). Since insulin biosynthesis and secretion was normal by virtue of the results of this thesis altered Ca2+ homeostasis may be an alternative/additional pathobiophysical mechanism that leads to type 1 diabetes mellitus in these mice. |
