Abstract: Der TRPM3-Ionenkanal gehört zur Familie der TRP-Kationenkanäle (TRP = "transient receptor potential"), die in einer Vielzahl von Geweben und Zelltypen exprimiert werden. Obwohl gewebsspezifische Liganden des TRPM3-Kanals derzeit unbekannt sind, kann dieser Ionenkanal durch das Neurosteroid Pregnenolonsulfat, den synthetischen Liganden CIM0216 und durch Temperaturen, die zu einer Verletzung des Gewebes führen können, aktiviert werden. In dieser Arbeit wurde ein HEK293-Zell-System verwendet, das es ermöglichte, die Expression des TRPM3-Ionenkanals selektiv durch die Gabe von Tetrazyklin zu induzieren. Durch die Verwendung dieses Systems wurde die durch die Agonisten Pregnenolonsulfat (PregS) und CIM0216 ausgelöste Signalkaskade und die darauffolgende Aktivierung verschiedener Transkriptionsfaktoren umfangreich charakterisiert. Die Stimulation von TRPM3 durch Pregnenolonsulfat steigerte die Aktivität der Transkriptionsfaktoren AP-1, CREB und Elk-1. In meiner Arbeit habe ich den Signalweg untersucht, der einen Ca2+-Einstrom nach der Aktivierung des TRPM3-Kanals mit einer erhöhten Expression des bZIP-Transkriptionsfaktors c-Fos, einem Zielgen von Elk-1, verbindet. Meine Analyse des Signalwegs zwischen aktiviertem TRPM3 und erhöhter c-Fos-Expression zeigte, dass sowohl MAP-Kinasen (ERK-Signalweg) als auch die Transkriptionsfaktoren CREB, SRF, AP-1, sowie Elk-1 Signalmoleküle sind, die an der Stimulus-induzierten Regulation der c-Fos- Gentranskription beteiligt sind. CIM0216, ein synthetischer Ligand des TRPM3-Ionenkanals, aktivierte den dimeren Transkriptionsfaktor AP-1 und stimulierte die transkriptionalen Aktivierungspotentiale von c-Jun und c-Fos. Es hatte jedoch ein geringeres Potenzial als PregS, die TRPM3-vermittelte Gentranskription zu aktivieren. Der Einstrom von Ca2+-Ionen in die Zellen ist von grundlegender Bedeutung für die Aktivierung von Elk-1 nach Stimulation von TRPM3. Ich konnte mithilfe von Calcium-bindenden Proteinen mit Zellkernlokalisationssequenz zeigen, dass ein Anstieg cytoplasmatischer Ca2+-Ionen für die Erhöhung des Transkriptionsaktivierungspotentials von Elk-1 entscheidend war, während die Pufferung von Ca2+ im Zellkern keine Wirkung auf die Transkriptionsaktivität von Elk-1 hatte. Die Untersuchungen ergaben weiter, dass die Stimulation der TRPM3-Kanäle zur Aktivierung der MAP-Kinase ERK (extrazelluläre signalregulierte Proteinkinase) führte, die wiederum eine wichtige Rolle bei der Stimulation der Transaktivierungsaktivität von Elk-1 führt. Weiterhin konnte ich herausfinden, dass die Ca2+/Calmodulin-abhängige Proteinphosphatase Calcineurin als ein negativer Rückkopplungsmechanismus der durch TRPM3 vermittelten Ca2+-abhängigen Stimulation von Elk-1 diente, da die Expression einer konstitutiv aktiven Calcineurin-Mutante im Zellkern zu einer verminderten Elk-1-Expression führte. Ein weiteres Ziel meiner Arbeit war es, durch den Transkriptionsfaktor AP-1 regulierte Zielgene in Folge der Stimulation des TRPM3-Ionenkanals zu finden. Dies führte zur Identifizierung des Interleukin-8 Gens (IL-8). Die Stimulation von TRPM3-Kanälen führte zu einer gesteigerten Aktivität des IL-8-Promoters. Eine eingehende Analyse des IL-8-Promotors mit verschiedenen Promotor-Mutanten ergab, dass die AP-1-Bindestelle des IL-8-Promotors eine entscheidende Rolle spielt. Zudem sind die basische Leucin-Zipper-Proteine c-Jun und ATF2 sowie der ternäre Komplexfaktor Elk-1 und die extrazelluläre signalregulierte Proteinkinase (ERK1/2) essentiell für eine verstärkte Transkription des IL-8-Gens nach TRPM3-Stimulation.
The TRPM3 ion channel belongs to the family of transient receptor potential (TRP) cation channels, which are expressed in a variety of tissues and cell types. Although tissue-specific ligands of the TRPM3 channel are currently unknown, this ion channel can be activated by the neurosteroid pregnenolone sulfate, the synthetic ligand CIM0216, and by moxious heat. In my thesis, a HEK293 cell system was used, which allowed to induce the expression of the TRPM3 ion channel selectively by the administration of tetracycline. By using this system, the signaling cascade triggered by the agonists pregnenolone sulfate (PregS) and CIM0216 and the subsequent activation of various transcription factors were extensively characterized. The stimulation of TRPM3 by pregnenolone sulfate increased the activity of the transcription factors AP-1, CREB and Elk-1. Here, I investigated the signaling pathway that links Ca2+ influx following activation of the TRPM3 channel with increased expression of the bZIP transcription factor c-Fos, a target gene of Elk-1. My analysis of the signaling pathway between activated TRPM3 and increased c-Fos expression showed that both MAP kinases (ERK signaling pathway) and the transcription factors CREB, SRF, AP-1, as well as Elk-1 are signaling molecules that are involved in the stimulus induced regulation of c-Fos gene transcription. CIM0216, a synthetic ligand of the TRPM3 ion channel, activated the dimeric transcription factor AP-1 and stimulated the transcriptional activation potentials of c-Jun and c-Fos. However, it had less potential than PregS to activate TRPM3-mediated gene transcription. The influx of Ca2+ ions into the cells is of fundamental importance for the activation of Elk-1 after stimulation of TRPM3. Using calcium-binding proteins with a nucleus localization sequence I could show that an increase in cytoplasmic Ca2+ ions was crucial for increasing the transcriptional activation potential of Elk-1, whereas buffering of Ca2+ in the nucleus had no effect on the transcriptional activity of Elk-1. The investigations further revealed that stimulation of the TRPM3 channels resulted in an activation of MAP kinase ERK (extracellular signal-regulated protein kinase), which in turn plays an important role in stimulating the transactivational activity of Elk-1. Furthermore, I found that the Ca2+/calmodulin-dependent protein phosphatase calcineurin served as a negative feedback mechanism of TRPM3-mediated Ca2+-dependent Elk-1 stimulation because expression of a constitutively active calcineurin mutant in the nucleus resulted in a decreased Elk-1 expression. Another goal of my work was to find AP-1-regulated target genes following stimulation of the TRPM3 ion channel. This led to the identification of the interleukin-8 gene (IL-8). Stimulation of TRPM3 channels resulted in an increased activity of the IL-8 promoter. A detailed analysis of the IL-8 promoter with different promoter mutants revealed that the AP-1 binding site of the IL-8 promoter plays a crucial role. In addition, the basic leucine zipper proteins c-Jun and ATF2 as well as the ternary complex factor Elk-1 and the extracellular signal-regulated protein kinase (ERK1 / 2) are essential for an enhanced transcription of the IL-8 gene following TRPM3 stimulation.
