Abstract: | The K1 toxin represents a heterodimeric A/B proteinaceous toxin that is secreted by Saccharomyces cerevisiae strains infected with two distinct double-stranded RNA viruses. In a two-staged receptor-mediated process, the ionophoric activity of K1 leads to an uncontrolled proton influx cumulating in the breakdown of the plasma membrane potential. In this study, various derivatives of the K1 precursor were constructed to analyse structural components essential for toxin activity as well as the mediation of functional immunity. Besides a further biochemical characterisation of the toxin, a transcriptome analysis of a sensitive strain after K1 toxin application was conducted revealing the significant up-regulation of various stress-regulating metabolic pathways, thereby, pointing not only to a complex toxic effect but also to a defence mechanism of non-killer yeast at least initially counteracting the ionophoric action. Furthermore, comparative analysis of two K1 killer strains with distinct phenotypical differences showed substantial intrinsic selection pressure in response to the level of toxin secretion. Moreover, the data suggest that immunity of the killer yeast is based on a balanced interplay between the toxin precursor within the secretory pathway and the number of toxin molecules attacking the toxin-producing cell. Das Killertoxin K1 stellt ein heterodimeres A/B-Toxin dar, welches von Stämmen der Bäckerhefe Saccharomyces cerevisiae sezerniert wird, die mit zwei spezifischen doppelsträngigen RNA-Viren infiziert sind. Der toxische Effekt des ionophoren K1-Toxins führt zu einem unkontrollierten Protoneninflux, der in einem Zusammenbruch des Plasmamembranpotentials kumuliert. In dieser Arbeit wurde durch Konstruktion unterschiedlicher K1-Derivate der Einfluss struktureller Komponenten auf die toxische Wirkung sowie die Vermittlung der Immunität charakterisiert. Neben einer weiterführenden biochemischen Charakterisierung des Toxins wurde mit Hilfe einer Transkriptomanalyse die zeitabhängige Reaktion eines sensitiven Stammes auf die Applikation des Ionophors analysiert. Hierbei wurde eine deutliche Induktion diverser zellulärer Stressreaktionen dokumentiert, die nicht nur auf einen komplexen Mechanismus des K1-Toxins, sondern auch auf eine initiale Abwehrreaktion sensitiver Zellen hindeuten. Weiterhin wurde mit Hilfe von K1-Killerstämmen die Co-Adaptation der Toxin-produzierenden Zellen in Abhängigkeit des Killervirus untersucht. Hierbei konnte die Ausbildung der Immunität als ein titrierbarer Mechanismus beschrieben werden, der einen distinkten intrinsischen Selektionsdruck auf Basis der Menge an sezerniertem Toxin ausübt. |
