Abstract: | Diese Doktorarbeit befasst sich mit der Totalsynthese und Konfigurationsrevision der Keramamide A und L sowie Mozamid A. Diese Naturstoffe stammen aus marinen Schwämmen der Gattung Theonella und gehören zur Familie der Anabaenopeptin-artigen Peptide. Die zentrale Herausforderung der Synthese war die Installation von Indol-Einheiten auf später Stufe ausgehend von stannylierten cyclischen Peptiden. Zunächst musste dazu eine Route zu N-Methylpropargylglycin enthaltenden Cyclopeptiden entwickelt werden. Der Organozinnrest konnte anschließend über eine Hydrostannylierung der Propargylgruppe eingeführt werden. Eine neuartige Sequenz aus Stille-Kreuzkupplung, Azidierung und photochemischer Nitren-C–H-Insertion führte zu den Tryptophan-haltigen Naturstoffen.
Nachdem die synthetische Methodik erfolgreich angewandt wurde, stellte sich durch sorgfältige Analyse der NMR-spektroskopischen Daten heraus, dass die ursprünlich angenommenen Strukturen von Keramamid A und L sowie Mozamid A inkorrekt zu sein schienen. Durch die Synthese von diastereomeren Strukturen konnte die eigentliche Konfiguration der Naturstoffe herausgefunden werden, sodass deren Strukturen revidiert werden konnten. Als proof of concept wurde anhand der neu entwickelten Methode der späten Indolsynthese eine Reihe diverser substituierter Keramamid-Derivate synthetisiert. This PhD thesis describes the first total syntheses and configurational revisions of Keramamides A and L as well as Mozamide A. These natural products were isolated from marine sponges of the genus Theonella and belong to the group of Anabaenopeptin-type peptides. The key challenge of the synthesis was the late stage installation of indole moieties on a stannylated cyclic peptide. Therefore, a route to N-methylpropargylglycine containing cyclic peptides was established. The organotin residue could then be introduced by hydrostannation of the popargylglycine-moiety. A novel sequence of Stille cross-coupling, azidation and photochemical nitrene C–H-insertion led to the tryptophan containing natural products. Having established this synthetic methodology, the originally proposed structures of Keramamides A, L and Mozamide A were found to be incorrect by careful analysis of the NMR-spectroscopic data. The synthesis of diastereomeric structures revealed the natural configurations of the natural products, whose structures could thus be revised. As a proof of concept, a series of diversely substituted Keramamide derivatives could be synthesized using the novel methodology. |
