Abstract: | Steroidhormone sind wichtige Regulatoren mannigfaltiger Vorgänge im Organismus. Ein Über- oder Unterangebot bestimmter Steroidhormone kann zu schwerwiegenden Krankheiten führen. Steroidogene Enzyme stellen daher interessante Zielmoleküle zur Therapie dieser Leiden dar. Die Hemmung der Androgenbildung ist ein erfolgreiches Therapiekonzept zur Behandlung hormonabhängiger Krankheiten wie dem Prostatakarzinom und dem benignen Prostatasyndrom. Diese Arbeit umfasst die Evaluierung der Hemmpotenz neuer Inhibitoren der androgenbildenden Enzyme 5α-Reduktase und CYP17A1. Neben etablierten Methoden kommt ein hier vorgestellter, neu entwickelter Assay zum Einsatz. Dieser ermöglicht die getrennte Hemmstoffbewertung gegenüber beiden Katalysereaktionen des bifunktionalen Enzyms CYP17A1 und bestätigt die These, dass zielgerichtete Hemmung des zweiten Katalyseschritts möglich ist. Weiter werden selektive Hemmstoffe jener Reaktion identifiziert und bilden die Grundlage zur Entwicklung nebenwirkungsarmer Wirkstoffe. Morbus Alzheimer ist eine neurodegenerative Krankheit, deren Ätiologie bislang nicht vollständig geklärt ist. Unzureichende Therapieoptionen machen die Notwendigkeit neuer Behandlungskonzepte deutlich. Eine Steigerung der Synthese zellschützender Neurosteroide könnte den Verlauf der Krankheit bremsen. In dieser Arbeit wird durch Versuche am Zellmodell gezeigt, dass die als neuroprotektiv beschriebene Gammahydroxybuttersäure keinen Einfluss auf die Aktivität steroidogener Enzyme hat. Steroid hormones are important regulators of manifold processes in the organism. Both, excess as well as insufficient supply of particular steroid hormones may lead to severe diseases. Therefore, steroidogenic enzymes represent interesting targets for the therapy of these illnesses. Inhibition of androgen formation is a successful concept of therapy for treating hormone dependant diseases like prostate cancer and benign prostatic syndrome. This thesis comprises the evaluation of the inhibitory potency of novel inhibitors of the androgen forming enzymes 5α-reductase and CYP17A1. Together with established methods, a newly developed assay is used. The latter allows a separate evaluation of inhibitors against either of the two reactions of the bifunctional enzyme CYP17A1 and confirms the theory that a targeted inhibition of the second catalytic step is possible. Moreover, selective inhibitors of this reaction are identified and provide the basis for the development of active pharmaceutical ingredients with low side effects. Alzheimer’s disease is a neurodegenerative disease with a not yet fully understood aetiology. Insufficient treatment options emphasise the need for novel therapeutic concepts. An increase of the synthesis of cell protecting neurosteroids may slow down the course of the disease. This thesis uses cellular model experiments to demonstrate that γ-hydroxybutyric acid, which is described as neuroprotective, has no influence on the activity of steroidogenic enzymes. |
