Abstract: | Das Ziel der vorliegenden Arbeit war die Entwicklung einer schritt-effizienten Synthese von Crocapeptin A, die für die Herstellung von Derivaten von Crocapeptinen verwendet werden kann. Darüber hinaus wurde eine Methode zur Modifikation von Di- bzw. Tripeptiden via allylischer Alkylierung entwickelt, die für eine "late-stage" Funktionalisierung zum Erhalt von Crocapeptin-analogen Verbindungen angewendet werden kann.
Die Synthesestrategie bezüglich des Depsipeptids zielte auf den Aufbau des cyclischen Kerns ab, an den anschließend die Einführung verschiedener Seitenketten möglich ist. In dieser Arbeit wurden zwei verschiedene Strategien zur Synthese des cyclischen Kerns bezüglich Ihrer Effizienz und Anwendbarkeit verglichen. Es konnte gezeigt werden, dass eine lineare Synthese zum Aufbau des Hexapeptids mit anschließendem Ringschluss der konvergenten Synthese vorzuziehen ist. Dies zeigte sich insbesondere in der Epimerisierungsempfinflichkeit der einzelnen Bausteine in der konvergenten Synthese.
Die Studien zur Peptidmodifizierung via allylischer Alkylierung am Di- bzw. Tripeptid zeigten deutlich, dass hydroxylierte und nicht-proteinogene Aminosäuren in den zu alkylierenden Peptiden die Reaktion limitieren. Schließlich konnte eine Methode entwickelt werden, bei der Allylglycin als Precurser für eine funktionalisierte Aminosäure diente. Dieser konnte im Anschluss der allylischen Alkylierung in das 5-Hydroxynorvalin überführt werden. The goal of this thesis was the development of a step-efficient synthesis of Crocapeptin A, that can be used for the production of derivatives of Crocapeptins. Furthermore, di- and tripeptides were modified via an allylic alkylation. This method can be applied for a late-stage functionalisation to obtain analogues of Crocapeptin. The strategy of the synthesis of this depsipeptide primarily focuses on the construction of the cyclic core. Afterwards, the introduction of different sidechains is possible. In this thesis two different synthetic strategies for the cyclic core are compared with respect to efficiency and universal applicability. It has been shown that the linear synthesis of the hexapeptide with a subsequent ringclosure is preferred to the convergent synthesis. This was indicated by epimerisation of several buildingblocks during peptidecouplings. The studies of peptidemodification towards di- and tripeptides via allylic alkylations unveil that non-proteinogenic, hydroxy-containing aminoacids can restrict the reaction. Ultimately, a method was developed in which allylglycin is used as a precurser for the functionalized sidechain, which can be transformed to the 5-hydroxynorvaline after the allylic alkylation. |
