Abstract: | Pyrrolobenzodiazepine sind DNA-alkylierende, nichtribosomal synthetisierte Sekundärmetabolite aus Actinomyceten. In dieser Arbeit wurden Studien zur Produktionsoptimierung von Tomaymycin sowie zur Darstellung von Tomaymycin und Sibiromycin-Varianten durchgeführt. Nach Isolierung des Tomaymycin- Biosynthese-Genclusters wurde eine Strategie zur heterologen Expression etabliert. Durch Optimierung der Vektor-Stamm-Kombination sowie genetische Manipulation wurde die Produktion im Vergleich zum Wildtyp um das siebenfache auf 340 mg/l gesteigert. Zudem wurde eine Mutasynthese-Strategie erarbeitete, mit welcher neuartige Tomaymycin-Varianten hergestellt wurden. Diese Methode ermöglicht die Biosynthese interessanter Ausgangsstoffe für zielgerichtete Wirkungsoptimierungen. Der ertragsmindernde Einfluss der Oxidation wurde mittels knock-out eliminiert, wobei in vivo ein neuer Abbaumechanismus, die Reduktion des Produkts, identifiziert wurde. Über heterologe Expression der NRPS der Tomaymycin-Biosynthese wurde ein funktioneller in vitro-Assay etabliert. In Studien zu einem weiteren PBD, Sibiromycin, wurden mittels vorläufergesteuerter Biosynthese neuartige, glykoslierte PBD-Varianten erzeugt, sowie ein System zur semisynthetischen Dimerisierung via Click-Chemie etabliert. Abschließend konnte eine Sibiromycin-Variante identifiziert werden, welche bei einer Dimerisierung eine deutlich erhöhte Toxizität aufweisen könnte; vergleichbar zum Dimerisierungseffekt von SJG-136. Pyrrolobenzodizepines are DNA alkylating, non-ribosomally synthesized, peptidic secondary metabolites found in actinomycetes. This work focuses upon optimization for the production of tomaymycine as well as mutasynthetic and precursor directed biosynthesis of new non-native variants of tomaymycine and sibiromycine respectively. For this purpose the tomaymycine biosynthetic genecluster was isolated and a heterologous expression system was established subsequently. By continuous vector/strain optimization and genetic enhancements production could be amplified sevenfold compared to the wildtype yielding a production titer of 340 mg/l. Moreover a strategy for mutasynthesis was built leading to several novel variants of tomaymycine. This newly developed method opens up new approaches for the biosynthesis of a broad spectrum of precursor substances and thereby facilitates directed lead optimization for drug effect and efficiency. The yield reducing oxidative effect was eliminated by knock-out experiments thus revealing a hitherto unknown in vivo degradation process, the reduction of the Pyrrolobenzodiazepine. A functional in vitro assay was designed via heterologous expression of the NRPS responsible for the biosynthesis of tomaymycine. Likewise studies on sibiromycine, another structurally related PBD, were performed via precursor directed biosynthesis and led to novel glycosylated PBD variants. By the use of a click-chemistry approach a system allowing for semisynthetic dimerization of PBD could be established. Eventually the presented studies provide a sibiromycine variant that once dimerized bears the potential for a highly boosted cytotoxicity - comparable to the effect observed for SJG-136. |
