Abstract: | In der vorliegenden Arbeit wird ein mikrofluidischer Zellkultur-Chip zur Optimierung von in vitro-Tests entwickelt. Auf dem Chip wird eine binär-logarithmische Verdünnungsreihe einer beliebigen Flüssigkeit durch ein System von Mikrokanälen passiv erzeugt, wodurch Kontaminationen minimiert und Untersuchungen zu Wechselwirkungen verschiedener chemischer Komponenten auf Zellen für viele Anwendungsbereiche ermöglicht werden, wie z. B. in der Medikamentenentwicklung und bei Toxizitätstests. Der Chipherstellung gingen analytische und numerische Berechnungen, die Auswahl der Herstellungsmethoden sowie die Entwicklung einer neuen Eckenkompensation für das anisotrope Ätzen voraus. Die Luftblasenbildung während des Befüllvorgangs wurde bei zwei unterschiedlichen SiO2-Oberflächenbeschichtungen untersucht. Eine neue Befüllmethode in der Unterdruckatmosphäre eines evakuierbaren Exsikkators wurde erfolgreich entwickelt. Außerdem wurde ein Versuchsaufbau zur Probengewinnung an jedem Well realisiert und geeignete Analysemethoden zur Ermittlung der Konzentrationen untersucht. Die Ergebnisse zweier Chipdesigns werden miteinander verglichen und die Funktionstauglichkeit der Siliziumchips wird durch Erfüllung dreier Kernfunktionen demonstriert: einer einheitlichen Flussrate an den Ausgängen, der Erzeugung einer binär-logarithmischen Verdünnungsreihe sowie der Kultivierung von lebenden Zellen in Kavitäten auf dem Chip bei kontinuierlicher Mediumzufuhr. In this work, a microfluidic cell culture chip is developed for optimization of in vitro tests. A binary logarithmic dilution profile of an arbitrary fluid is passively generated on such a chip by a system of microchannels, reducing contaminations and enabling a broad range of applications by analyzing the interaction of different chemical compounds on cells--for example, in drug development and toxicity tests. The manufacturing process was preceded by analytical and numerical calculations, a selection of manufacturing methods as well as the development of a new convex corner compensation for anisotropic etching. Formation of air bubbles during the filling process of the chip was analyzed with respect to two different SiO2 surface coatings. A new filling method was developed successfully, using the low pressure environment in a vacuum desiccator. Furthermore, a test setup for extracting one sample per well was engineered, and suitable methods of analysis for measuring the concentrations were investigated. The results of two chip designs are compared with each other, and the functionality of the silicon chip is demonstrated by achieving three core functions: a uniform flow rate at the outlets, the generation of a binary logarithmic dilution gradient, and the cultivation of living cells inside on-chip cavities with continuous media perfusion. |
