Abstract: | Diese Dissertation beschreibt die Totalsynthese von Cyclomarin C, einem Vertreter einer Familie von marinen Naturstoffen mit einzigartiger biologischer Aktivität gegenüber Tuberkulose-auslösenden Bakterien. Es handelt sich dabei um ein komplexes, cyclisches Heptapeptid, welches neben drei proteinogenen Aminosäuren aus vier unnatürlichen besteht, von denen zwei exklusiv in dieser Naturstofffamilie vorkommen. Zur Synthese dieser Teilbausteine wurden neue, hochselektive und gleichzeitig kurze Routen entwickelt. Neben Cyclomarin C, für das bereits zuvor eine Totalsynthese beschrieben war, konnten zwei weitere Vertreter dieser Naturstoffklasse, Cyclomarin A und D, erstmals synthetisiert werden. Darüber hinaus wurde ein vereinfachtes Derivat hergestellt, welches durch eine fehlende Hydroxygruppe eine erheblich kürzere Synthese erfordert. Interessanter Weise besitzt es die gleiche antituberkulotische Aktivität im unteren mikromolaren Bereich wie die beiden potentesten Naturstoffe Cyclomarin A und C. This PhD-work describes the total synthesis of Cyclomarin C, a marine natural product with a unique biological activity against tuberculosis-causing bacteria. It is a complex, cyclic heptapeptide, which consists of three natural and four non proteinogenic amino acids, of which two occur exclusively in this family of natural products. For the synthesis of these building blocks, highly selective and straightforward routes have been developed. Besides Cyclomarin C, for which a total synthesis was already described, two more members of this natural product class, Cyclomarin A and D, were synthesized for the very first time. Furthermore, a simplified derivative, which requires a shorter reaction sequence due to a missing hydroxyl group, was synthesized. Interestingly, it possesses the same antituberculotical activity as the two most potent natural products Cyclomarin A and C in the lower micromolar range. |
