Abstract: | Die vorliegende Arbeit untermauert die Hypothese, dass es in Erythrozyten zwei unterschiedliche Wege zur PS-Exposition geben muss. Durch Vergleich verschiedener Methoden (Anregung zur PS-Exposition und Erhöhung des intrazellulären Ca2+-Gehaltes durch A23187, LPA und PMA sowie Inhibierung von Scramblase, PKC und Gardos-Kanal) war es möglich, Unterschiede im Reaktionsverhalten der Zellen zu analysieren. Es wurde deutlich, dass sich das Reaktionsverhalten der Zellen klar unterscheidet, wenn verschiedene Mechanismen aktiviert werden. Hierbei steht der durch A23187 und LPA aktivierte Signalweg dem durch PMA aktivierten Signalweg gegenüber.
Weiter wurden Parameter, die in der Literatur als mögliche Trigger für die PSExposition diskutiert werden, analysiert. Der Efflux von K+-Ionen konnte, zumindest im durch A23187 und LPA induzierten Signalweg, als für die PS-Exposition mitverantwortlicher Faktor identifiziert werden. Im PMA-induzierten Signalweg hingegen spielt er keine Rolle. Eine Änderung des Zellvolumens sowie eine Alkalisierung des intrazellulären pH-Wertes konnten als Trigger für die PS-Exposition ausgeschlossen werden.
Es wurde der Nachweis erbracht, dass die Auftrennung der Erythrozyten in unterschiedliche Altersklassen mittels Percoll-Separation verlässliche Ergebnisse liefert. Dass die Unterschiede im Reaktionsverhalten der Erythrozyten einer Population im unterschiedlichen Zellalter begründet sein könnte, kann jedoch ausgeschlossen werden und muss andere Ursachen haben. It is known that erythrocytes have 2 different pathways leading to PS exposure. The presented work strongly supports this hypothesis. To analyse differences in their reactions behavior cells were treated under different experimental conditions (PS exposure and influx of extracellular Ca2+ were stimulated using A23187, LPA and PMA. Furthermore, Scramblase, PKC and Gardos channel were inhibited). The results show that activation of the different pathways leads to diverse cellular responses: the signal pathway activated by A23187 and LPA is different from the pathway activated by PMA. In literature, different parameters are taken into consideration for PS exposure (K+ efflux, changes in cell volume and alkalization of intracellular pH). These parameters were analysed in the presented work, too. The results show that an efflux of K+ is involved in the mechanism of PS exposure in the case of the A23187- and LPAactivated pathway. However, K+ efflux does not play a role in the PMA-activated pathway. Changes in cell volume and alkalization of intracellular pH can be excluded as possible causes for PS exposure. Separation of erythrocytes into fractions of different cell age was performed. It was proven that the method gives reliable results. The presented data clearly show that the cell age is not responsible for these differences. In addition, erythrocytes from one donor react different after stimulation for PS exposure. |
