Abstract: | Die vorliegende Arbeit befasst sich mit der Synthese von Cyclodextrin-Polymeren und der Solubilisierung lipophiler Wirkstoffe. Im ersten Teil wurden native und methylierte Mono-Azid-Cyclodextrine (CD) über „Click“-Chemie an propargylierte Hydroxyethylstärke (HES) gekuppelt. Durch Einführen von Carboxylatgruppen an HES oder die Kupplung an Hyaluronsäure (HA) konnte das Auftauchen einer LCST vermieden werden. Zudem wurde gezeigt, dass die gekuppelten methylierten CDs eine signifikant geringere Zytotoxizität als die entsprechenden Monomere aufweisen. Der zweite Teil behandelt die Amidkupplung nativer und methylierter Mono-Amino-CDs an HA und oxidierte HES mit CDMT als Kupplungsreagenz. Im dritten Teil wurden in einer zweistufigen Synthese methylierte CDs mit H-Phosphonat monofunktionalisiert und mit Pivaloylchlorid als Aktivierungsreagenz an Hydroxypropylcellulose gekuppelt. Im letzten Teil wurde das Einschlussverhalten der synthetisierten CD-Derivate auf die lipophilen Wirkstoffe Sevofluran, Midazolam, Propofol, Hydrocortison, Estradiol, Testosteron und Ibuprofen mit Hilfe von ITC, der Methode der Löslichkeitsisothermen und der Headspace-Gaschromatographie untersucht. Die Ergebnisse zeigten einen deutlich stärkeren Solubilisierungseffekt auf die Wirkstoffe als natives β-CD und seine kommerziellen Derivate. Die vorgestellten, sehr gut wasserlöslichen CD-Polymere stellen deshalb vielversprechende Kandidaten für die Entwicklung neuer Darreichungsformen lipophiler Wirkstoffe dar. This thesis discusses the synthesis of new cyclodextrin (CD) polymers and the complexation of hydrophobic drugs. In the first part mono-azides of native and methylated CDs were conjugated to propargylated hydroxyethyl starch (HES) by “click”-chemistry. The appearing LCST phase separation could be avoided completely by the introduction of carboxylate groups or the use of hyaluronic acid (HA) as a polymer backbone. The conjugated methylated CDs exhibited significantly lower cytotoxicity than the corresponding monomeric CD derivatives. The second part describes the conjugation of mono-amino functionalized CDs to HA and oxidized HES using CDMT as a coupling agent. For the resulting water soluble polymers no cytotoxic effect could be detected. In the third part a two-step synthesis is presented in which CDs were functionalized by H-phosphonate and were conjugated to hydroxypropyl cellulose using pivaloylchloride as a condensing agent. In the last part the inclusion behaviour of these CD-derivatives with the poorly soluble drugs sevoflurane, midazolam, propofol, hydrocortisone, estradiol, testosterone and ibuprofen were investigated using the phase solubility method, ITC and headspace gas chromatography. The studies demonstrated higher solubilizing abilities of the CD polymers in comparison to commercially available β-CD derivatives. The CD polymers with both low toxicity and excellent binding potential are promising candidates for new dosage forms of hydrophobic active ingredients. |
