Abstract: | Die Familie der Chondramide wurde im Jahr 1995 im Zuge eines Naturstoff-screenings als Sekundärmetabolite in Kulturextrakten der myxobakteriellen Spezies Chondromyces crocatus gefunden. Strukturell bestehen die 18-gliedrigen Cyclodepsipeptide aus einer Tripeptid- [L-Ala, N-Methyl-D-Trp (D-Abrin) und L-β-Tyr] und einer Polyketideinheit. Die Chondramide verfügen über eine beeindruckende cytostatische Wirkung gegenüber diversen Krebszelllinien, indem sie aktinregulierende Zellvorgänge stören und als Konsequenz den Zelltod einleiten.
Diese Doktorarbeit beschreibt die Synthese neuer Chondramid-Derivate mit unterschiedlichen Substitutionsmustern im Polyketidsegment sowie mit modifizierten Peptidsequenzen aus stellenweise halogenierten Aminosäuren. In Zusammenarbeit mit Prof. Dr. R. Müller (Universität des Saarlandes) wurde anhand dieser kleinen Substanzbibliothek ein Beitrag zum Verständnis der Struktur-Aktivitäts-Beziehungen geleistet. Dadurch konnten der Einfluss der verschiedenen Reste abgeschätzt, die strukturelle Komplexität der Chondramide bei Aktivitätserhalt abgebaut und der Zugang zu wirksamen Derivaten durch Vereinfachung der Synthese erleichtert werden. Zudem konnte ein fluoreszenzmarkiertes Chondramid hergestellt werden, um durch „live cell imaging“- und „co-localization“-Studien den Wirkmechanismus und mögliche weitere biologische Angriffsziele identifizieren zu können. The chondramides were found as secondary metabolites in culture extracts of myxobacteria species Chondromyces crocatus in 1995 by a natural product screening. The structure of the 18-membered cyclodepsipeptides consists of a tripeptide [L-Ala, N-methyl-D-Trp (D-Abrin) and L-β-Tyr] and a polyketide subunit. The chondramides have an impressive cytostatic effect on various cancer cell lines by interrupting actin-regulating cellular processes. This work describes the synthesis of new chondramide derivatives with different substitution patterns of the polyketide subunit and modified peptide sequences of partially halo-genated amino acids. Owing to the cooperation with Prof. Dr. R. Müller (Saarland University) a deeper understanding of the structure-activity-relationships of this small substance library has been achieved. Thereby, the influence of the various residues was estimated and the structural complexity of the chondramides was decreased while maintaining the activity. The access to effective derivatives was facilitated due to the simplified synthesis of the compounds. Furthermore, a fluorescently labeled chondramide was prepared to identify the biological mechanism of action and further potential targets by „live cell imaging”- and „co-localization“-studies. |
