Abstract: | Die vorliegende Arbeit behandelt die templatgesteuerten Synthesen von tubularen α-Cyclodextrinsystemen:
Der erste Teil beschreibt die Herstellung von tubularen α-Cyclodextrin Dimeren. Hierzu wurde zunächst ein Bis-1,20-(2-Methylpyridinium)eicosan Templat aus α,ω-Dibromeicosan und 2-Picolin synthetisiert. Daraus wurden [3]-Pseudorotaxane auf Basis von zwei α-CDs und dem kationischen Bola-Amphiphil hergestellt. Die komplexierten α-CDs wurden anschließend in basischer Lösung mit Epichlorhydrin verknüpft. Abschließend wurde das Templat in heißem DMSO entfernt und die Dimere mittels Ionenaustauschchromatographie isoliert.
Im zweiten Teil der Arbeit wurden Untersuchungen zur Herstellung von organischen „molecular tubes“ (MT) per Olefinmetathese durchgeführt. Hierzu wurden Polyrotaxane auf Basis von α-Cyclodextrin und Polyethylenglykol 10.000 hergestellt und die Syntheserouten im Bezug auf Ausbeute und Belegungsgrad optimiert. Zudem wurde gefunden, dass durch Ca(SCN)2 eine bis zu zehn prozentige Steigerung des Belegungsgrades, in Abhängigkeit zur Salzkonzentration, beim Auffädelprozess der Polyrotaxane erreicht wird. Die Polyrotaxane wurden in einem LiCl/DMAc Gemisch hochgradig (89 %) mit Allylgruppen substituiert. Abschließend wurde untersucht, ob sich die Olefinmetathese (Grubbs 2) zum Verknüpfen der funktionalisierten Polyrotaxane eignet, was zu Umsätzen von 20 % aller Allylgruppen führte. The present thesis discusses the template-directed synthesis of tubular α-cyclodextrin systems: The first part describes the synthesis of tubular α-cyclodextrin dimers. On this a bis-1,20-(2-methylpyridinium)eicosane template was synthesized out of α,ω-dibromoeicosane and 2-picoline, in order to prepare a [3]-pseudorotaxane, based on two α-CDs and this cationic bolaamphiphile. Following the complexed α-CDs were crosslinked by epichlorhydrin in an alkaline solution. After removing the template in hot DMSO solution, the cyclodextrin dimers were finally isolated by ion exchange chromatography. The second part of this thesis discusses the research of synthesizing molecular tubes (MT). Based on α-cyclodextrin and polyethylene glycol 10.000, polyrotaxanes were prepared and its routes optimized for yield and coverage. Furthermore, using Ca(SCN)2 during the threading process was found, depending on its concentration, to increase coverage of polyrotaxanes by ten percent. Using LiCl/DMAc solutions, the polyrotaxanes were functionalized by etherification with allyl bromide in a high degree of substitution (89 %). Finally olefin metathesis (Grubbs 2 catalyst) was examined for crosslinking these modified polyrotaxanes, leading to conversions of 20 % of all allyl groups. |
