Abstract: | Im Rahmen der vorliegenden Arbeit wurden verschiedene Pipecolinsäure-Derivate synthetisiert, die von der Arbeitsgruppe von Prof. R. Müller am Institut für Pharmazeutische Biotechnologie zur Untersuchung der Biosynthese des antibiotischen Naturstoffs GE81112 eingesetzt wurden. Dabei fungierten die hergestellten Verbindungen entweder als Referenzen für die zu analysierenden Naturstoffbausteine oder sie wurden für Umsetzungen mit an der Biosynthese von GE81112 beteiligten Enzymen verwendet. Im zweiten Teil der Arbeit wurde ein Amidenolat aufgebaut, das zur Palladium-katalysierten intramolekularen allylischen Alkylierung geeignet ist. Zunächst wurde ermittelt welche Elemente diese Allylierungsvorstufe beinhalten muss, um eine möglichst effiziente Cyclisierung zu ermöglichen. In diesem Zusammenhang wurde dann ein entsprechendes Substratspektrum entworfen und Chancen sowie Limitierungen der allylischen Substitution ausgetestet. Durch die gewonnenen Erkenntnisse konnten wiederum optimierte Vorstufen synthestisiert werden, die schließlich zu noch besseren Ergebnissen bei der allylischen Alkylierung führten. Within the scope of the present thesis different derivatives of pipecolic acid were synthesized and subsequently applied by the group of Prof. R. Müller at the Department of Pharmaceutical Biotechnology for examinations concerning the biosynthesis of the antibiotic GE81112. Thereby the synthezised compounds were either used as referents for those building blocks of the natural product to be analyzed or they were applied for the enzymatic conversion by such enzymes being engaged in the biosynthesis of GE81112. In the second part of the thesis an amide enolate was built up to be suitable for a palladium catalyzed intramolecular allylic alkylation. First, all those elements of the allylation precursor were explored which are necessary for a most efficient cyclization. In this context a corresponding substrate spectra was created and scope and limitation of the allylic substitution were tested. By the resulting information some optimized allylation precursors could be synthesized finally leading to even better results in the allylic alkylation. |
