Abstract: | Nanomaterialien haben das Potential, in vielfältigen biomedizinischen Applikationen eingesetzt zu werden. Die vorliegenden Ergebnisse zeigen jedoch deutlich auf, dass verschiedene Nanopartikel und Materialien neben ihrem gewünschten Effekt auch unspezifische Effekte an Erythrozyten und anderen Zelltypen auslösen können. Bei der Analyse der verschiedenen Nanopartikel wurden bei SiO2- und Au-Nanopartikeln keine Effekte gefunden, jedoch beeinflussen nackte Fe3O4-Nanopartikel den pH-Wert von Erythrozyten. Die Fe3O4-Nanopartikel, welche im Rahmen des EU-Projektes NanoMagDye hergestellt wurden und in der medizinischen Applikation eingesetzt werden sollen, zeigten keine Toxizität und Beeinflussung der untersuchten Zelltypen. Somit sollten diese Nanopartikel in der humanen Anwendung unbedenklich sein, da sie keine toxischen Nebeneffekte auslösen. Im Gegensatz dazu wurde bei fluoroalkylierten Phosphaten, welche zur Synthese von Mikrobläschen verwendet werden, ein deutlicher Anstieg der Hämolyse gefunden. Da die Mikrobläschen selbst aus Stabilitätsgründen nicht untersucht werden konnten, muss gewährleistet werden, dass diese bis zur fertigen Anwendung eine ausreichende Beständigkeit besitzen. Nanopartikel sind innovative Werkzeuge zur biomedizinischen Verwendung, jedoch muss bei potentiellen Anwendungen eine genauere Betrachtung von Nebeneffekten und toxischen Auswirkungen durchgeführt werden. Nanomaterials have a high potential of usage in many different biomedical applications. The presented results show clearly that different nanoparticles and materials show unspecific effects on erythrocytes and other cell types. The analysis of different nanoparticles presents no effects for SiO2 and Au nanoparticles. In contrast naked Fe3O4 nanoparticles affect the intracellular pH for erythrocytes. The nanoparticles produced in the EC project NanoMagDye which are planned to be used in the medical application show now toxicity and influence on the investigated cell types. Therefore these nanoparticles should be harmless in the human application. On the other side we found a significant increase in the rate of haemolysis after the incubation with fluoroalkylated phosphates (used for synthesis of microbubbles). For stability reasons, we could not analyse the microbubbles themselves. Therefore it must be ensured that the microbubbles have a adequate stability before used in the application. Nanoparticles are innovative tools for biomedical applications, but we have to ensure for potential applications, which toxic side effects can occur. |
