Abstract: | CYP11B2 katalysiert die Aldosteron–Synthese von DOC über B und 18OHB. Da CYP11B2 Mutationen die Ursache für Aldosteronsynthase–Defizienz sind, liefern Studien zu Mutationen und ihren Folgen auf die Enzymaktivität wertvolle Beiträge für die Diagnose und Therapie von Hypoaldosteronismus. In dieser Arbeit wird die Entwicklung eines schnellen und preiswerten Ganzzellsystem zur enzymatischen Analyse von CYP11B2 Mutanten vorgestellt. Das Prinzip beruht auf der heterologen Expression der Mutanten in S. pombe, gefolgt von der Analyse der CYP11B2–abhängigen Biotransformation von DOC. Nach Optimierung der Systemparameter wurden zehn CYP11B2 Mutationen untersucht. Vergleiche mit aus Zellkulturstudien gewonnenen Literaturdaten bestätigen die Eignung des Hefesystems zur Analyse der Effekte von CYP11B2 Mutationen. Somit stellt das etablierte System eine innovative, kostengünstige und Radioaktivitätsunabhängige Alternative zur Charakterisierung von CYP11B2 Mutanten dar. Des Weiteren wurde mithilfe des zuvor etablierten Systems das biotechnologische Potential von S. pombe als 18OHB–Produzent untersucht. Hiefür wurden weitere 18 CYP11B2 Mutanten untersucht. Es musste jedoch festgestellt werden, dass neben der noch optimierungsbedürftigen Enzymaktivität ein weiterer limitierender Faktor darin besteht, dass 18OHB in wässrigem saurem Milieu dazu neigt, Umlagerungsprodukte zu bilden. Daher muss gefolgert werden, dass Mikroorganismen zum jetzigen Wissensstand zur 18OHB–Produktion ungeeignet sind. CYP11B2 catalyses the formation of aldosterone from DOC via B and 18OHB. Mutations in the CYP11B2 gene cause aldosterone synthase deficiency, so studies of their effect on the enzyme activity provide useful information for the diagnosis and therapy of hypoaldosteronism. Here, the development of a fast and cost–efficient whole–cell system for the enzymatic characterisation of CYP11B2 mutants is described. The approach is based on the heterologous expression of the mutants in S. pombe combined with a subsequent biotransformation of DOC. After optimising the system parameters, ten CYP11B2 mutations were investigated. A comparison of the obtained results with previously described results from cell culture experiments confirmed the suitability of the yeast system for further studies on CYP11B2. Thus, an innovative, low–cost and non–radioactive alternative for the characterisation of CYP11B2 mutants was successfully established. Furthermore, the biotechnological potential of S. pombe as 18OHB producer was assessed using the system described above. Therefore, another 18 CYP11B2 mutants were investigated. However, it was discovered that the biotechnological applicability of the system is severely limited not only by the unfavourable enzymatic activity, but also by the structural rearrangements 18OHB undergoes in aqueous acidic medium. Due to the latter point it has to be concluded that at least currently microorganisms seem to be generally unsuitable for 18OHB production. |
