Abstract: | Diese Dissertation beschreibt die Synthese von vereinfachten Derivaten des Tubulysin D-Biosyntheseintermediates Prätubulysin, wobei die zentrale Aminosäure Tubuvalin, durch die relativ einfachen Bausteine Oxazoltubuvalin, Phenyltubuvalin, Phenoxytubuvalin und Triazoltubuvalin ersetzt wurde. Diese neuen Aminosäuren können in wenigen Schritten aus leicht zugänglichen Ausgangsmaterialien gewonnen werden. Die neuen Prätubulysin-Derivate, namentlich Oxazolprätubulysin, Phenylprätubulysin, Phenoxyprätubulysin und Triazolprätubulysin, sind zwar weniger aktiv im Vergleich zu den natürlichen Tubulysinen, dennoch liegt die Aktivität einiger Derivate gegenüber diverser Tumorzellen weiterhin im nanomolaren Bereich. Darüber hinaus wurde Hydroxyprätubulysin synthetisiert, um die letzten Geheimnisse der Tubulysin-Biosynthese aufzuklären. Dieses Projekt läuft in Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. R. Müller vom Institut für Pharmazeutische Biotechnologie an der Universität des Saarlandes. Außerdem beschäftigt sich die Arbeit mit der Synthese von photoaktivierbaren Prätubulysin-Derivaten zur Identifikation neuer biologischer Angriffsziele der Tubulysine. Der Kooperationspartner ist in diesem Fall die Arbeitsgruppe von Herrn Prof. Dr. S. A. Siebert von der LMU München. This PhD-work describes the synthesis of simplified derivatives of pretubulysin, a biosynthetic intermediate of tubulysin D, where the central amino acid, tubuvaline, is replaced by the rather simple building blocks oxazoletubuvaline, phenyltubuvaline, phenoxytubuvaline and triazoletubuvaline. These new amino acids can be obtained in few steps from easily accessible starting materials. The new pretubulysin derivatives, called oxazolepretubulysin, phenylpretubulysin, phenoxypretubulysin and triazolepretubulysin are less active compared to the natural tubulysins, but the activity of several derivatives against different tumor cells are still in the nM range. Furthermore hydroxypretubulysin was synthesized to confirm some speculations of the tubulysin-biosynthesis in collaboration with the group of Prof. Dr. R. Müller (Department of Pharmaceutical Biotechnology, Saarland University). In collaboration with the group of Prof. Dr. S. A. Siebert (LMU Munich), the thesis deals with the synthesis of photoreactive pretubulysin derivatives to identify new targets of tubulysins. |
