Abstract: Das viral kodierte, antimykotisch wirksame Killertoxin Zygocin der Hefe Z. bailii konnte in aktiver Form ohne Aktivitätsverlust bis zur Homogenität gereinigt werden. Kryo-EM Untersuchungen negativ geladener PC/PA-Liposomen zeigten nach Inkubation mit gereinigtem Zygocin eine vollständige Zerstörung, neutral geladene Liposomen dagegen blieben intakt. Analytische Ultrazentrifugationsanalysen zygocinbehandelter, hydrophober Detergensmizellen zeigten eine Oligomerisierung des Toxins in den Mizellen. Es kann daher angenommen werden, dass Zygocin in vivo Plasmamembranporen ausbildet, vermutlich aus vier Untereinheiten bestehend. Zygocin zeigte eine geringe Löslichkeit und neigte bei hohen Konzentrationen zur Präzipitatbildung. Eine Plasmid-getriebene Expression und Sekretion von biologisch aktivem Zygocin in P. pastoris wurde erfolgreich etabliert. Das Toxin wies eine deutlich höhere Aktivität auf als das natürlicherweise vom Wildtypkillerstamm produzierte Virustoxin. Die Hochzelldichtefermentation von P. pastoris zur Produktion von aktivem Zygocin und mRFP-Zygocin konnte etabliert werden. Das postulierte Modell zur ionophoren Toxinwirkung konnte durch die in vivo Expression verschiedener Toxinvarianten und deren Phänotypanalyse verifiziert werden. Deletionen der amphipathischen α-Helix sowie der Transmembranhelix führten ebenso wie einzelne Punktmutationen zu einem vollständigen Aktivitätsverlust des Toxins. Überraschenderweise führte die Deletion der bislang für die Primärrezeptor-Erkennung als relevant postulierten D1-Domäne zu einem unverändert aktiven Toxin, so dass angenommen werden kann, dass die geringe Größe des Toxins (4,1 kDa) auch ohne Bindung des Mannoprotein-Rezeptors die Überwindung der Zellwand erlaubt.
The viral encoded antimycotic killertoxin zygocin of the yeast Z. bailii could be purified to homogeneity and in an active form. Cryo-EM analysis showed a complete destruction of negatively charged PC/PA-liposomes after incubation with purified zygocin. In contrast, neutral loaded liposomes stayed intact. Oligomerization of the toxin could be observed via analytical ultracentrifugation analysis of zygocin treated hydrophobic detergent micells. Based on this observation, it can be suggested that zygocin builds pores containing four subunits in the plasma membrane in vivo. Zygocin possesses a low solubility and tends to precipitate at higher concentrations. The plasmid-driven expression and secretion of biological active zygocin in P. pastoris was established yielding a significant higher activity compared to the toxin of the wild-type killer strain. Further, production of both active zygocin and mRFP-zygocin in P. pastoris could be established using high cell density fermentation. The postulated model of zygocin's ionophoric effect could be verified in vivo by expression of different toxin variants and phenotypical analysis of these variants. Deletion of the amphipathic α-helix or the transmembrane helix as well as single point mutations led to a complete loss of toxin activity; the same was true for single point mutations. Surprisingly, deletion of the D1-domain, which should be necessary for the primary receptor recognition, showed no negative effect on activity. Therefore, it could be assumed that the small size of the toxin (4,1 kDa) allows the overcoming of the cell wall even without binding the mannoprotein receptor.
