Abstract: | Die makrocyclischen Bisibibenzyle - Marchantine, Plagiochine, Riccardine und Isoplagiochine - sind Naturstoffe, die ausschließlich in Moosen vorkommen. Totalsynthesen einiger Verbindungen dieses Strukturtyps sind bereits bekannt. Im Rahmen dieser Arbeit wurden neue Strategien zur enantioselektiven Synthese der Isoplagiochine entwickelt. Hierzu wurden kinetische Racematspaltungen via Reaktion der Stilbendoppelbindung sowie Übergangsmetall katalysierte Cyclisierungen untersucht. Im Verlauf der Arbeit wurde zum einen Bisbibenzyl-Makrocyklen via Ringschlussmetathese aufgebaut, zum anderen gelang ein atropselektiver Ringschluss durch intramolekulare asymmetrische HECK-Reaktion. Weiterer Gegenstand dieser Arbeit war die Totalsynthese biologisch aktiver Bisbibenzyle des Plagiochin- und Riccardin-Typs. Diese wurde gemäß einer einheitlichen Syntheseroute durchgeführt. Die detaillierte spektroskopische Auswertung der Produkte führte zu einer Strukturrevision des isolierten, biologisch hochaktiven Plagiochin E, das aufgrund seiner antitumoralen Wirkungen potentieller Kandidat für ein neues Anti-Krebs-Therapeutikum ist. Bei der stereochemischen Untersuchung der synthetisierten Verbindungen konnte - erstmalig für Verbindungen des Plagiochin- und Riccardin-Typs - das Vorliegen von Atropisomeren nachgewiesen werden. Macrocycles of the bisbibenzyl-type - marchantins, plagiochins, riccardins and isoplagiochins - are natural products that are found exclusively in bryophytes. The total syntheses of several bisbibenzylic compounds have already been reported. In the course of this work new strategies for an enantioselctive synthesis were developed. For this purpose kinetic resolutions via reactions at the stilbene double bond as well as transition metal catalyzed cyclisations were investigated. During this work, the ring-closure of bisbibenzylic macroycles was achieved via ring-closing metathesis as well as an atroposelective ringclosure applying an intramolecular asymmetric HECK-reaction. Another concern of the work was the total synthesis of bioactive bisbibenzylic compounds of the plagiochin- and riccardin-type. The syntheses were established by a general approach. The detailed spectroscopic analysis necessitated a structure revision of the isolated, highly bioactive plagiochin E, which represents a potential cancer therapeutic agent due to its antitumor activities. During the stereochemical investigations of the synthesized compounds the existence of atropoisomers could - for the first time - be verified for compounds of the plagiochin- and riccardin-type. |
