Abstract: Das Enzym Aldosteron-Synthase (CYP11B2) katalysiert die letzten Schritte der Biosynthese des Aldosterons. Die Hemmung dieses Enzyms stellt ein vielversprechendes Therapiekonzept zur Behandlung von Erkrankungen dar, bei denen erhöhte Aldosteronkonzentrationen im Plasma eine Rolle spielen, wie z.B. bei einer Herzinsuffizienz oder einer Myokardfibrose. 3-Pyridylsubstituierte Naphthaline sind potente CYP11B2-Inhibitoren, die jedoch das hepatische Enzym CYP1A2 hemmen und nicht in der Lage sind in vivo (Ratte) den Aldosteronspiegel zu senken. Zur Optimierung dieser Hemmstoffe wurden Substituenten an C-1 und C-2 des Naphthalins eingeführt. Dabei wurde ein neuer synthetischer Zugang zu 1,3 disubstituierten 2-Naphtholen entwickelt, die sich als potente und gegenüber den Enzymen CYP11B1, CYP17 und CYP19 hochselektive CYP11B2-Inhibitoren herausstellten. In einem zweiten Ansatz führte die Modifikation des mit dem Pyridylrest substituierten Rings des Naphthalins durch Einführen von Heteroatomen und polarer Gruppen zu einem Verlust der inhibitorischen Wirkung auf das Targetenzym CYP11B2. Im letzten Teil dieser Arbeit wurden ausgehend von pyridylsubstituierten Dihydrochinolinonen neue hochpotente Inhibitoren des humanen CYP11B2-Enzyms gewonnen, die gegenüber dem hoch homologen CYP11B1 selektiv sind und darüber hinaus in der Lage sind in vitro die Aldosteron-Synthase der Ratte zu hemmen.
Aldosterone synthase (CYP11B2) catalyses the last steps in aldosterone biosynthesis. Therefore, the inhibition of this enzyme offers a promising therapy concept for the treatment of cardiovascular diseases associated with chronically elevated aldosterone plasma levels. Examples include hypertension, congestive heart failure and myocardial fibrosis. 3-Pyridine substituted naphthalenes are potent CYP11B2 inhibitors. However, they are not selective towards the hepatic enzyme CYP1A2 and they are not able to reduce aldosterone plasma levels in vivo (rat). Optimization of these compounds by introduction of substituents at C-1 and C-2 of the naphthalene core resulted in the development of a new synthetic approach to 1,3-disubstituted 2-napthols, which turned out to be potent CYP11B2 inhibitors showing excellent selectivities towards CYP11B1, CYP17, and CYP19. In a second approach the pyridyl substituted ring of the naphthalene was modified by introduction of heteroatoms and polar groups resulting in a loss of inhibitory potency towards the target enzyme CYP11B2. In the last part of this work, using pyridyl substituted dihydroquinolinones as lead structure, new highly potent human CYP11B2 inhibitors were developed that show good selectivities towards the highly homologous CYP11B1 and are able to inhibit the rat CYP11B2 in vitro.
