Abstract: Die nicht proteinogene Aminosäure Nicotianamin (NA) ist ein in höheren Pflanzen ubiquitärer und essentieller Chelator von Schwermetallen. In Gramineen fungiert NA als direkte biochemische Vorstufe in der Synthese der Phytosiderophore und ist damit indirekt an der Aufnahme von Fe und Zn in die Wurzel beteiligt. Wie die NA freie, semi-sterile Tomatenmutante chloronerva zeigt, erfüllt NA auch in Nicht-Gramineen eine essentielle Funktion, die bislang jedoch nicht genau verstanden ist. Mit zunehmender Verbreitung der Modellpflanze Arabidopsis thaliana ergaben sich vielfältige neue, für Tomate nicht verfügbare Untersuchungsmethoden, die einen Transfer der Erforschung des NA Mangels auf A. thaliana für langfristige Studien attraktiv machten. In der vorliegenden Arbeit wird die Generierung und Charakterisierung einer auf T-DNA Mutagenese beruhenden und die vier Nicotianamine Synthase (NAS) Gene betreffenden Mutantenkollektion in A. thaliana beschrieben. Die Analyse der fertilen nas4x-1 Mutante ergab eine geringe NAS Restaktivität, die sich in deutlich verringerten, aber detektierbaren NA Gehalten äußerte. Dennoch wurden umfassende Veränderungen in den Metallgehalten sowie der Genexpression bekannter, in der Metallhomöostase involvierter Gene beobachtet, wodurch eine bessere Integration der NAS Gene in das Netzwerk der Metallhomöostase ermöglicht wurde. Die durchgeführten Analysen deuteten insbesondere auf eine essentielle Funktion des NA in der Entwicklung der reproduktiven Organe hin. Darüber hinaus wurden Hinweise auf eine essentielle Funktion des NA in der Toleranz gegenüber dem Schwermetall Nickel sowie gegenüber Eisenmangel gewonnen. NA ist demnach essentiell sowohl in der Ernährung als auch in der Detoxifizierung von Metallen. Darüber hinaus erfüllt es essentielle Aufgaben in der vegetativen und reproduktiven Phase der Pflanzenentwicklung.
The non-proteinogenic amino acid nicotianamine (NA) is essential for chelation and transport of iron and other micronutrient metal ions in higher plants. In graminceous plants, NA is the direct precursor in the synthesis of phytosiderophores and thus directly involved in the acquisition and uptake of essential metal ions from the soil. However, due to the analysis of the NA free and semi-sterile tomato mutant chloronerva, an essential role of NA also in non-graminaceous plants is generally accepted. With the establishment of Arabidopsis thaliana as the major dicot model plant many new methods were developed which are not available in Solanaceous species. Thus, a change of the model species to investigate loss of NA in A. thaliana, became attractive. Due to the NAS gene family structure in A. thaliana multiple nas mutants were constructed, enabling the identification of intermediate nas mutant phenotypes to study NA production and transport during reproduction. The nas4x-1 mutant, being mutated at all four NAS genes, contained drastically reduced but still detectable amounts of NA, enabling the mutant to sustain its life cycle. Anyway, global changes in metal homeostasis and the expression of known metal homeostasis genes were detected in the nas4x-1 mutant, leading to a better understanding and integration of the NAS genes into the network of metal homeostasis. The obtained results strongly indicate an essential role of NA in the development of flowers and seeds. Furthermore, it was shown that an increasing number of nas mutations leads to a higher sensitivity against high amounts of nickel as well as iron deficiency. Thus, NA plays essential roles in metal nutrition and metal detoxification both at vegetative and reproductive stage of plant development.
