Abstract: | Diverse Krankheitsbilder, wie Diabetes mellitus, Arthritis oder Krebs, gehen häufig mit einer fehlerhaften Regulation von Matrix-Metalloproteasen, insbesondere MMP-2 und MMP-9, einher. Die Validierung neuartiger Hemmstoffe wird vor allem durch den hohen Kostenaufwand bei der Isolierung der benötigten MMPs aus Tumor-Zelllinien oder Fibroblasten erschwert. Vor diesem Hintergrund wurde in der vorliegenden Arbeit ein zellbasierter MMP-Bioassay für ein Hochdurchsatz-Screening nach potentiellen Inhibitoren etabliert, der auf der heterologen Expression und Immobilisierung von MMP-2 und MMP-9 auf der Zellwand von Hefen basiert. Zur Identifikation eines geeigneten Expressionswirts wurden die humanen Gelatinasen in den Hefen K. lactis, P. pastoris, S. cerevisiae, S. pombe und Z. bailii zur Sekretion gebracht. Dabei war P. pastoris als einziger Organismus in der Lage, beide MMPs in ausreichend hoher Qualität als biologisch aktive Enzyme zu produzieren. Im Rahmen vergleichender Experimente zur Zellwand-Expression in P. pastoris konnte mit Sed1p aus S. cerevisiae ein hoch effizientes, neuartiges Zelloberflächen-Expressionssystem für P. pastoris identifiziert werden, das die für S. cerevisiae maximal erreichbare Immobilisierungs-Effizienz um den Faktor 6,7 übertrifft. Die Anpassung eines kommerziell erhältlichen Gelatinase-Assays an die Anforderungen eines zellbasierten Systems ermöglichte einen quantitativen Vergleich der Hemmstoff-Wirkungen durch ein einfaches, standardisiertes Messverfahren. A wide range of diseases, such as diabetes mellitus, arthritis and, most of all, cancer, goes along with malfunctions in the regulation of matrix metalloproteinases, especially MMP-2 and MMP-9. The invention and validation of novel inhibitors is often slowed down by the high costs of MMP purification from human tumor cell lines or primary fibroblasts. In view of this background, an innovative cell based MMP bioassay was invented to enable a high throughput screening for potential MMP inhibitors. This method is based on the heterologous expression and immobilization of MMP-2 and MMP-9 on the cell surface of yeast. To identify an appropriate expression host, the human gelatinases were expressed as secretory proteins in the yeasts Kluyveromyces lactis, Pichia pastoris, Saccharomyces cerevisiae, Schizosaccharomyces pombe and Zygosaccharomyces bailii. P. pastoris appeared to be the only organism capable of producing both MMPs in sufficiently high quality. Comparative studies on cell surface expression considering S. cerevisiae cell wall proteins revealed Sed1p as being a novel and highly efficient carrier system for cell surface display in P. pastoris that exceeded immobilization efficiency of S. cerevisiae by the factor 6.7. A commercial gelatinase assay was adapted to match the requirements of a cell based system, thus providing an efficient tool for quantification of MMP inhibitor activities via a simple, standardized bioassay. |
