Abstract: | Die pi-sigma-pi-Isomerisierung von terminalen pi-Allyl-Komplexen ist ein entscheidender limitierender Faktor für die Synthese (Z)-konfigurierter Substitutionsprodukte und immer noch ein weitgehend ungelöstes Problem in der Palladium-katalysierten allylischen Alkylierung. In der Regel isomerisiert die cis-konfigurierte Doppelbindung zur thermodynamisch stabileren trans-Doppelbindung. Bis jetzt sind erst wenige Beispiele von Übergangsmetall-katalysierten Reaktionen unter Erhalt der (Z)-Olefingeometrie bekannt. Im Rahmen dieser Arbeit konnte eine Methode entwickelt werden, die es erlaubt cis-Allylsubstrate mit chelatisierten Glycin-Esterenolaten unter Palladium-katalysierten Bedingungen ohne Verlust der Doppelbindungsgeometrie umzusetzen. Durch Verwendung optisch aktiver Substrate konnte überdies eine effizienter 1,5-Chiralitätstransfer in das neu geformte Aminosäurezentrum erreicht werden. Verschiedene alkyl- und arylsubstituierte gamma,delta-ungesättigte Aminosäuren konnten auf diese Weise mit guten cis-Selektivitäten sowie Diastereoselektivitäten erhalten werden. Das erarbeitete Verfahren konnte insbesondere seine synthetisch wertvolle Anwendung in der stereoselektiven Synthese C-glycosidisch verknüpfter Aminosäuren beweisen. Weiterhin ermöglichte die Entwicklung einer isomerisierungsfreien Palladium-katalysierten allylischen Substitution, mechanistische Details wie den Memory-Effekt näher zu untersuchen. The pi-sigma-pi-isomerization of terminal pi-allyl complexes is a decisive limiting factor of the synthesis of (Z)-configured substitution products and is to a great extent an unsolved problem in the palladium catalyzed allylic alkylation due to the isomerization of the cis-configured double bond to the thermodynamically favoured trans-double bond. Only few examples of the transfer of the cis-olefin geometry under transition-metal-catalyzed conditions have been reported so far. This thesis deals with the evolution of a method that enables a palladium-catalyzed reaction of (Z)-allylic substrates with chelated glycine ester enolates without loss of the double bond geometry. By use of optical active compounds an efficient 1,5-chirality transfer to the new formed stereogenic amino acid center could be observed and a variety of alkyl- and arylsubstituted gamma,delta-unsaturated amino acids could be obtained in a highly stereoselective fashion. In particular, this approach found application in the stereoselective synthesis of C-glycosidic amino acids. Furthermore, the development of an isomerization-free palladium-catalyzed procedure allowed the investigation of mechanistic details such as the memory effect. |
