Abstract: Ein Teilaspekt der vorliegenden Arbeit sollte die Induzierbarkeit des Amplifikationsprozesses innerhalb der vorhandenen Amplifikationseinheit 12q13-14 nachweisen. Dazu wurde die Glioblastomzelllinie TX3868 mit Aphidicolin oder mini-Hypoxie behandelt und die dadurch induzierten Auswirkungen bezüglich des Amplifikationsprozesses analysiert und dargestellt. Hierzu zählen die Induktion von double minutes variabler Größe, Brüche der HSR-Strukturen und die Reintegration von double minutes. Dabei konnte eine bisher noch nicht beschriebene common fragile site in unmittelbarer Nähe des Gens KUB3 identifiziert werden. In dem zweiten Teilprojekt der vorliegenden Arbeit sollte der Einfluss verschiedener Zellkulturbedingungen auf neurale Tumorstammzellen untersucht werden. Die Existenz von Tumorstammzellen in Gehirntumoren konnte in früheren Studien nachgewiesen werden. Des Weiteren zeigt sich beim Glioblastom eine Resistenz gegenüber Röntgenstrahlung. In der vorliegenden Arbeit konnte durch Verwendung spezieller Medien, sowie der Reduktion des Sauerstoffpartialdruckes (mini-Hypoxie), die Bildung von Neurospheres, wie sie typisch für neurale Stammzellen ist, induziert werden. Darüber hinaus konnte die Fusion von Neurospheres mittels Immunfluoreszenzfärbung nachgewiesen werden. Nach 15 Gy Bestrahlung wurde in den TX3868-Zellen ein 11,50 %-iger Anstieg neuraler Tumorstamm-zellen festgestellt. Eine Bestrahlung mit 40 Gy führte zu einer Steigerung der neuralen Tumorstammzellpopulation um 9,98 %. Die hier aufgezeigten Resultate verdeutlichen, dass die konventionelle Therapieform des Glioblastoms, mit dem Ziel der Zerstörung der malignen Zellpopulation, die Ausbildung gegenteiliger Effekte bedingen kann. Die Bestrahlung der Glioblastomzellen führte zu einer Anreicherung neuraler Tumorstammzellen, aus denen vermehrt neue Tumorzellen gebildet werden können.
One aspect of the present work should provide evidence that amplification can be induced within the existing amplification unit 12q13-14. For that purpose, glioblastoma cell line TX3868 was incubated with aphidicolin and mini hypoxia. The induced effects of the amplification process were analyzed and described. There were double minutes of variable size, breakages of hsr structures and reintegration of double minutes. Furthermore, a so far not described, common fragile site could be identified, which is localized in proximate distance to the gene KUB3. The second part of the present work should regard the influence of various cell culture conditions on glioblastoma derived stem cells. Former studies could demonstrate the existence of tumor stem cells in brain tumors. Furthermore, glioblastomas show resistance against irradiation. In the present work, generation of neurospheres that is typical for neural stem cells could be induced by application of different media and the reduction of oxygen partial pressure (mini hypoxia). Moreover, fusion of neurospheres was detected using immunostaining. Furthermore, irradiation of TX3868 cells lead to increasing levels of neural tumor stem cells after radiation dosages of 15 Gy (+ 11.50 %) and 40 Gy (+ 9.98 %). The major objective of the standard therapy of glioblastomas is the elimination of malignant tissue. However, the accumulation of neural tumor stem cells after irradiation of glioblastoma cells leads probably to increased levels of tumor cells that have been arisen out of tumor stem cells.
