Abstract: Aufgrund der immer noch hohen Zahl an offenen Fragenstellungen in den Biowissenschaften gilt die Proteomanalyse als eine der wichtigsten Forschungsgebiete in der Bioanalytik, welche sich intensiv mit der Aufklärung der Gesamtheit aller Proteine der Zelle beschäftigt. Auf dem Weg zum Lösen der Fragestellungen spielt die Schnelligkeit und die Zuverlässigkeit der ausgewählten Analysenmethode eine große Rolle. Aufgrund der geringen Menge an verfügbarem Probenmaterial werden häufig miniaturisierte chromatographische Trenntechniken wie Mikro-HPLC innerhalb der biowissenschaftlichen Forschung eingesetzt, welche mit verschiedenen massenspektrometrischen Detektionstechniken kombiniert werden. In der vorliegenden Arbeit wurde ein Elektrospay-Ionisation-(ESI)-Ionenfallen- Massenspektrometer sowohl für die qualitativen als auch für die quantitativen Analysen verwendet, welches sich auch als sehr empfindliche und schnelle Detektionsmethode herausstellte. Im ersten praktischen Teil der Arbeit wurden Gliadine identifiziert, die zu den Glutenproteinen gehören. Die erste Art der Identifizierung wurde mit Hilfe der intakten molekularen Massen der Gliadine durchgeführt. Zu diesem Zweck wurde eine Tuningmethode durch Verwendung von Cytochrom-C optimiert. Mit dem optimierten System wurden unter 17 verschiedenen Peaks insgesamt 41 Proteinmassen ermittelt. Daraus wurden zwei a/ß-Gliadine, ein a-Gliadin und drei γ-Gliadine identifiziert. Da die zumal mit der Genauigkeit einer Ionenfalle - gemessene Proteinmasse keinesfalls ausreichende Informationen zur Proteinidentifizierung liefert, wurden als zweiter Weg die Proteine chymotryptisch und peptisch verdaut. Anschließend wurden die resultierenden Peptide mittels Tandem-MS durch Peptidfragmentfingerprinting (PFF)identifiziert. Mit Hilfe einer Kombination der Suchmaschinen Mascot, InsPect, OMSSA und deren Algorithmen wurden die Peptide identifiziert. Zur Identifizierung konnten diese sechs Gliadine eindeutig nach ihrer Gliadinfamilie zugeordnet werden. Eine eindeutige Identifizierung dieser sechs Gliadine war jedoch aufgrund der Homologie nicht möglich. Im zweiten Teil der vorliegenden Arbeit wurde eine Quantifizierungsmethode entwickelt, mit der ein toxisches 30-mer Peptid quantifiziert wurde. Das toxische 30-mer Peptid wurde durch peptischen Verdau in Gegenwart einer besonderen aktiven Denaturierungssubstanz (RapiGestTM) aus einem Weizenextrakt gewonnen. Dabei wurden die Verdaubedingungen hinsichtlich der Verdauzeit optimiert. Entsprechend dieser Optimierungsstudien wurden vier parallele peptische Verdaue zur Bestimmung der Reproduzierbarkeit und zur Quantifizierung durchgeführt. Die Kalibrierung für die absolute Quantifizierung wurde mit einem isotopenmarkierten internen Standard vorgenommen. Als interner Standard wurde ein Peptid verwendet, welches dieselbe Sequenz des toxischen 30-mer Peptides hatte und am N-terminalen Valin durch sechs C-13-Atome isotopenmarkiert war. Der letzte Schritt dieser Quantifizierungsstudie bestand in der Auswertung der Rohdaten nach einem Feature-Finder-Algorithmus mit dem Ziel, dass durch die Automatisierung der Auswertungsprozesse eine schnelle Datenauswertung erreicht werden kann. Zur Kontrolle der automatischen Auswertung wurde eine manuelle Auswertung der Daten durchgeführt. Dabei wurde eine gute Übereinstimmung der Ergebnisse beider Auswertungsprozesse festgestellt. Um diese Quantifizierungsmethode in der alltäglichen Praxis der Lebensmittelindustrie zu realisieren, wurde anschließend zu diesem Zweck das toxische 30-mer Peptid in einem kommerziell erhältlichen Weizenmehl von Typ 405 erfolgreich qualitativ und quantitativ untersucht.
Due to the still remaining high number of unanswered questions in life sciences, the proteome analysis is currently considered as one of the most important research tools of bioanalysis. In general, proteome analysis deals with identification, structural studies and posttranslational modification studies of the whole cell proteome. Hereby the rapidity and reliability of the selected analysis method are some of the most challenging issues. Due to the usually small sample amounts to be analyzed, a miniaturized chromatographic separation technique such as micro-HPLC combined with mass-spectrometric detection techniques is frequently used. In the present work an electrospray ionization (ESI) ion trap mass spectrometer was used for the qualitative and quantitative analyses. The method represented itself as a very sensitive and fast detection method. In the first part of the present work gliadins were identified, which belong to the gluten proteins. The first way of identification was accomplished by measuring the molecular masses of the intact gliadins. For this purpose a tuning-method could be optimized using cytochrome c. With this optimized system, 17 different peaks were determined, which represented a total number of 41 protein masses. From these 41 protein masses two a/ß-gliadins, one a-gliadin and three γ-gliadins were identified. To confirm these results, a second method for protein identification using peptic and chymotryptic digestions of proteins, Tandem-MS and Peptide Fragment Fingerprinting (PFF) was applied. The identified peptides were used to determine their corresponding proteins using a combination of the search machines Mascot, InsPect, OMSSA and their algorithms. Six gliadins could be clearly assigned to their gliadin family. A doubtless identification of these six gliadins was not possible due to their structural homology. In the second part of the present work, a quantification method was developed to quantify a toxic 30-mer peptide. The toxic 30-mer peptide was isolated from a wheat extract by peptic digestion in presence of a special denaturing agent (RapiGestTM). The digest conditions were optimized with respect to the digestion time. According to the optimization studies four parallel peptic digests were prepared for quantification and reproducibility study purposes. An isotope-labeled internal standard was used for the calibration. A synthetic 30-mer toxic peptide containing six C-13 atoms in the N-terminal valine was used as an internal standard. The last step in this quantification study consisted of the analysis of the raw data applying the Feature Finder algorithm to achieve a quick data analysis by automating the evaluation process. A manual data analysis was also performed to validate the results of the automatic data evaluation. The results of both the manual and the automatic data analysis showed to be in very good agreement. The project then was finalized by applying this optimized analysis method to commercially available type 405 wheat flour where this toxic 30-mer peptide was analysed successfully qualitatively and quantitatively.
