Abstract: | Die Funktionalisierung von Elektrodenoberflächen mit Biomolekülen spielt sowohl für analytische als auch präparative Anwendungen eine wichtige Rolle. Viele Enzyme, wie die Galaktit- und Sorbitol-Dehydrogenase (GatDH / SDH), benötigen zur Substratumsetzung das Cofaktorsystem NAD+/NADH, um ein Substrat zu oxidieren. Die elektrochemische Regenerierung des Cofaktors stellt eine Alternative zum klassischen Recycling dar und ermöglicht die enantiomerenreine Bildung eines Produktes. Hierzu wurden alle benötigen Bestandteile wie Redoxmediator, Cofaktor und Enzym auf die Elektrodenoberfläche immobilisiert und die elektrochemische Regenerierung des Cofaktor mittels cyclischer Voltammetrie untersucht. Des Weiteren wurde auch die elektrochemische Regenerierung des Cofaktors an einer mit Cystein modifizierten GatDH-Variante untersucht, welche durch Ausbildung einer Thiolatbindung direkt auf die Elektrode immobilisiert wird. Die Pyranose-2-Oxidase (P2Ox) katalysiert die regioselektive Oxidation von Polyhydroxyaldehyden zu Ketoaldosen, ermöglicht den Zugang zu pharmazeutisch relevanten Zuckerderivaten und wurde ebenfalls immobilisiert. Zur Erhöhung der aktiven Oberfläche wurden nanokristalline Elektroden mittels Pulstechnik bzw. durch galvanostatische Abscheidung hergestellt. Ferner sollten Platin-Inseln auf Au (111)-Oberflächen mit definiertem Abstand durch eine Kombination aus stromloser Abscheidung und Elektroabscheidung hergestellt werden, um eine anschließende Immobilisierung der P2Ox zu ermöglichen. Functionalisation of electrode surfaces with biomolecules is important for analytical and preparative applications. Enzymes like galaktit- and sorbitol dehydrogenase(GatDH / SDH) need the cofactor system NAD+/NADH in order to oxidise a substrate. The electrochemical regeneration of this cofactor describes an alternative to the classical recycling and allows the preparation of an enantiomerically pure product. For this reason all components were immobilized on the electrode surface as redox mediator, cofactor and enzyme and the electrochemical regeneration of the cofactor was investigated by cyclic voltammetry. In addition the electrochemical regeneration of the cofactor was investigated at a GatDH modified with cysteine which was immobilized on the electrode via the formation of a thiolat bond. Pyranose-2-oxidase (P2Ox) catalyses the regioselective oxidation of polyhydroxyaldehyde to an ketoaldose, permits the access to important pharmaceutical sugar derivatives and was also immobilized. Nanocrystalline electrodes prepared by pulse techniques or by galvanostatic deposition were used to enhance the active surface. Furthermore platinum islands should be produced on Au (111) by a combination of electroless and electrochemical deposition to enable a subsequent immobilisation of P2Ox. |
