Abstract: Die vorliegende Arbeit behandelt die gezielte Synthese von wasserlöslichen Cyclodextrinderivaten, die an der Position C-6 mit kationischen oder anionischen Gruppen substituiert sind sowie die systematische Untersuchung der Stabilität ihrer Einschlussverbindungen mit anionischen, kationischen und neutralen tert.-Butylbenzolderivaten mittels isothermer Titrationskalorimetrie. Es wurden mittels dreier Methoden sowohl mono- als auch heptasubstituierte ß Cyclodextrinderivate synthetisiert. Bei Methode A wurde OH-6 durch eine mehrstufige Synthese in eine Aminogruppe überführt, von der aus weitere Modifikationen erfolgten. In Methode B wurden Substituenten über eine Thioetherbrücke an C-6 angeknüpft. Die Cyclodextrinderivate waren ohne Schutzgruppen und unter Erhalt der freien Hydroxylgruppen am Cyclodextrin mit hoher Regioselektivität zugänglich. In einer dritten Methode C wurden aminosäuremodifizierte Cyclodextrine regioselektiv in hoher Ausbeute erhalten. Dazu wurden reaktive Azido-Cyclodextrinderivate mit propargylierten Aminosäuren in einer kupferkatalysierten 1,3-dipolaren Cycloaddition umgesetzt. Um den Einfluss des pH-Wertes und der Salzkonzentration auf die Stabilität der Einschlussverbindungen zu untersuchen, wurden die Titrationen bei pH 3 und pH 7 in 25 mM und 500 mM Phosphatpuffer durchgeführt. Alle Einschlussverbindungen zeigten eine 1:1 Stöchiometrie. Die Bindungskonstanten der Monoderivate bewegten sich im Bereich von 2000 M-1 und 54000 M-1, abhängig von der Kombination der funktionellen Gruppen von Wirt und Gast und der Pufferkonzentration. Die Erkennung zwischen den funktionellen Gruppen kann hauptsächlich Coulomb-Wechselwirkungen und Wasserstoffbrückenbindungen zugeschrieben werden. Es zeigte sich, dass schon im nativen ß-Cyclodextrin eine natürliche Ladungserkennung vorhanden war, die unter anderem vom Dipolmoment des -Cyclodextrins und von der Hydrophilie der Gäste abhing. Im Allgemeinen wurden anionische Gäste stärker gebunden als kationische Gäste. Es wurde beobachtet, dass die Ladungsdiskriminierung bei Mono-(6-deoxy-6-guanidyl)-ß-cyclodextrin am stärksten ausgeprägt war und bei Mono-(6-deoxy-6-S-thioglykolat)-ß-cyclodextrin Natriumsalz fast vollständig verschwand. Die Orientierung des Gastes innerhalb der Kavität des Cyclodextrins wurde stark von den Wechselwirkungen zwischen den funktionellen Gruppen von Wirt und Gast beeinflusst, jedoch spielten auch Dipol-Dipol-Wechselwirkungen eine nicht unerhebliche Rolle. ROESY-Experimente haben gezeigt, dass sich der Gast in der Kavität immer so orientiert, dass eine antiparallele Ausrichtung der Dipolmomente von Wirt und Gast die Folge ist. Bei den heptafunktionalisierten Cyclodextrinen wurden im Vergleich zu den Monoderivaten wesentlich höhere Stabilitätskonstanten beobachtet. In einem Fall lagen diese erstaunlicherweise mit einem Wert von 5.9•106 M-1 in einer Größenordnung, die für monomere Cyclodextrine bislang nicht beobachtet wurde. Verbindungen, die in der Lage sind Komplexe in dieser Stärke zu bilden, sind für viele pharmazeutische Anwendungen vorstellbar. Damit konnte die Löslichkeit des Zytostatikums Camptothecin durch Zusatz von Per-(6-cysteaminyl-6-deoxy)-ß-cyclodextrin auf mehr als das Zehnfache gesteigert werden. Somit gibt diese Arbeit neue Impulse zum Verständnis der molekularen Erkennung von Cyclodextrinen.
The present thesis discusses the selective synthesis of water-soluble derivatives of cyclodextrins, substituted with cationic or anionic groups at the C-6 position as well as the systematic investigation of the inclusion of anionic, cationic and neutral derivatives of tert.-Butylbenzene by these water-soluble cyclodextrin derivatives by isothermal titration calorimetry (ITC). Mono- as well as hepta-derivatives of cyclodextrine were synthesized. The preparation of these derivatives included three methods. Method A was the conversion of the OH-6 to an amino-group. This amino-group was the starting point for further modifications. Method B described the attachment of the substituent by a thioether bridge at C-6. All derivatives were accessible with high regioselectivity and without protecting groups as well as conservation of the free hydroxyl groups. Furthermore strategy C was developed to obtain amino acid modified cyclodextrins in high selectivity and yield. In this strategy reactive azide derivatives of cyclodextrin undergo conversion with propargylated amino acids in a copper catalyzed 1,3 dipolar cycloaddition. The influence of the pH value and the salt concentration on the inclusion compound stability was investigated by measurement of the binding constant at pH 3 and pH7 in 25 mM and 500 mM buffer concentration. All inclusion compounds showed 1:1 stoichiometry while the binding constants of the mono derivatives ranged from 2000 M-1 to 54000 M-1 depending on the combination of the functional groups at host and guest as well as the buffer concentration. Recognition between functional groups was mainly attributed to Coulomb interactions and hydrogen bonds. Already native ß-cyclodextrine shows some intrinsic recognition potential towards charged guests, which is due to the total dipole moment of  cyclodextrine and differences in the hydrophilicities of anionic and cationic compounds. In general, anionic guests were stronger bound than cationic guests. We found that charge discrimination is largest in the case of Mono-(6-deoxy-6-guanidyl)-ß-cyclodextrine and on the other hand in the case of Mono-(6-deoxy-6-S-thioglycolate)-ß-cyclodextrine sodium salt the charge recognition no longer exists. Besides the coulombic interaction that mainly attributed to this behavior, however the dipole-dipole interactions play also an important role. Orientations of the guests within the cavity of the cyclodextrins were determined by ROESY NMR. In these experiments we found that orientations of the guest ever lead to antiparallel direction of the dipole moments of host and guest. The hepta-derivatives show significant higher binding constants compared to the mono-derivatives. In one case, we found a stability constant with a value of 5.9•106 M-1 which is in the order of magnitude of cyclodextrine-chelate complexes. Compounds with these high binding constants are interest for many pharmaceutical applications. For example, the solubility of the anticancer agent Camptothecin was ten times higher by adding a solution of Heptakis-(6-cysteaminyl-6-deoxy)-ß-cyclodextrine. The complete work furnishes new impulses to the understanding of the molecular recognition of cyclodextrins.
