Abstract: | Ziel der Arbeit war der Entwurf und die Etablierung einer hoch flexiblen und high-throughput faehigen Strategie zur Vorhersage intestinaler, passiver Absorption, welche dazu in der Lage ist, den Arzneimittelentwicklungsprozess von Anfang bis zum Ende zu begleiten. Diese Strategie ist dazu in der Lage, sich der bestehenden Situation anzupassen und finazielle und menschliche Ressourcen, aber auch Zeit und die Vervuegbarkeit von Substanz zu beruecksichtigen.
Des weiteren konnte eine neue und innovative Methode zur Vorhersage der Permeabilitaet entwickelt werden, die auf der Bestimmung zweier Lipophilieparameter, logP(Oct) und logP(PGDP), beruht.
Letztenendes wurde die etablierte Strategie an einem konkreten Beispiel eingesetzt, der Verbesserung der Permeabilitaet eines vielverprechenden und hoch potenten 5-alpha Reduktase Inhibitors. The goal of this work was to develop and to establish a high flexible and high-throughput strategy for passive permeability prediction, capable to support the drug development process from the very beginning to it` s end. The strategy is able to adopt on the actual situation and facts like financial and human resources, time and the availability of drug substances. Furthermore an innovative, new permeability prediction method was established, basing on the determination of two lipophilicity parameters, logP(Oct) and logP(PGDP). Finally the established strategy was used to improve the permeability of a promising, high potent 5-alpha reductase inhibitor. |
