Abstract: | Fehlregulationen von Histondeacetylasen (HDACs) sind entscheidende Ereignisse der Pathogenese der akuten myeloischen Leukämie (AML) und die Basis für innovative, epigenetische Behandlungsstrategien, zu denen besonders der Einsatz von HDAC-Inhibitoren zählt. Durch die AML-Subtypspezifischen Genexpressionsanalysen konnten Unterschiede im Expressionsmuster der HDACs aufgedeckt und AML M1 als Subtyp mit signifikant höheren HDAC Expressionen identifiziert werden, was Einfluss auf die Wirkung und Anwendung von HDAC-Inhibitoren haben könnte und einen zusätzlichen diagnostischen Marker darstellen könnte (eingereicht: Dransfeld, et al. 2010).
Die Analyse nicht synonymer SNPs (nsSNPS) in Klasse I HDACs mit putativer Funktion bei Anfälligkeit für AML deckte nsSNPs als Falschpositive auf und zeigten, dass sie in den hoch konservierten HDACs in Kaukasiern nicht existieren (Dransfeld, et al 2007). Die Analyse der zwei validierten nsSNPs von SIRT3, welches in allen analysierten AML Proben als hochreguliert aufgedeckt wurde, zeigte, dass sie keinen Einfluss auf Aktivität, zelluläre Funktionen im Energiemetabolismus oder die SIRT3 Stabilität, für deren Halbwertszeit diese Studie erste Hinweise lieferte, aufweisen (Dransfeld, et al).
Im Kontext der Optimierung und Individualisierung der Ara-C Therapie konnte mit dem Cytidin Deaminase SNP C-451T ein neuer, signifikanter, unabhängiger prognostischer Überlebens-Marker für AML Patienten aufgedeckt werden (Mahlknecht, et al 2009). Missregulation of Histone deacetylases (HDACs) is considered as a crucial event in acute myeloid leukemia (AML) pathogenesis and therefore the basis for innovative epigenetic treatment strategies, whereby HDAC-inhibitors takes part of these new therapeutic strategies. With our AML subtype specific gene expression analyses of HDACs we could reveal subtype specific HDAC expression patterns, with AML M1 representing a subtype with significant higher HDAC expression levels, which could hardly affect the impact of HDACi-therapy and present an additional diagnostic marker of AML subtypes (submitted: Dransfeld, et al. 2010). Concerning putative functions of class I HDAC nsSNPs in susceptibility to AML our analyses showed that there actually do not exist any class I HDAC nsSNPs in Kaukasians (Dransfeld, et al 2007). By analysing two nsSNPs of SIRT3, which was over expressed in all analysed AML probes, it was shown that they had no influence on SIRT3 activity, cellular functions in energy metabolism or stability, which was defined for the first time in this study (Dransfeld, et al). Finally, in the context of optimization of Ara-C standard therapy, this work leads to the identification of cytidine deaminase SNP C-451T as a new, significant, independent prognostic factor for survival of AML patient treated with Ara-C, which further prompts the need for individualized adjusted chemotherapy in patients who are being given Ara-C or other nucleoside analogs (Mahlknecht, et al 2009). |
