Abstract: Durch Ablösen einzelner Untereinheiten mit Hilfe des Detergens Lauryldimethyloxid (LDAO) konnten aus der V1ATPase (A3:B3:C:D:E:F:G2:Hy) aus Manduca sexta hydrolytisch aktive Subkomplexe gewonnen werden. Diese waren A3B3DEG (176% Hydrolyseaktivität relativ zur V1ATPase), A3B3HEGF (53 %), A3B3EG (58 %) und A3B3DEGH (47 %). Der zur Hydrolyse minimal notwendige A3B3EG-Subkomplex wurde hinsichtlich der Lage der Stieluntereinheiten E und G elektronenmikroskopisch untersucht. Die Bildanalyse der mit Uranylacetat negativ kontrastierten A3B3EG-Moleküle zeigte die sechs hexagonal und alternierend angeordneten Untereinheiten A und B. Im Zentrum des Hexamers befand sich die Untereinheit E mit G. Der A3B3EG-Subkomplex wurde auch im Verhalten beim zeitaufgelösten proteolytischen Verdau mit der vollständigen V1ATPase aus M. sexta und der Untereinheit E aus der Hefe Saccharomyces cerevisiae verglichen. Während die ungeschützte Untereinheit E aus S. cerevisiae innerhalb weniger Minuten gespalten wurde, verzögerte der partielle Schutz durch die katalytischen Untereinheiten A und B im A3B3EG-Subkomplex die Spaltung der Untereinheit E durch Trypsin. In der kompletten V1ATPase war die Untereinheit E durch die Stieluntereinheiten C, D, F, G und H von der Proteolyse abgeschirmt, so dass die Spaltung noch weiter protrahiert wurde. Der zur Topologiebestimmung genutzte Nulllängenvernetzer CuCl2 führte zu den Vernetzungsprodukten A—B—E, A—B—D—E und D—E. Die Kreuzvernetzung mit dem photoaktivierbaren Nukleotidanalogon P1,P4-Di-(3';-Biotinyl-8-azidoadenosin-5';-)-tetraphosphat (8-DiN3-DiB-AP4A) ergab die Produkte E—G, A—F(—G), A—B—E—G und D—E. Das mit beiden Kreuzvernetzern auftretende Produkt aus dem zentralen Kopplungselement E und der Stieluntereinheit D zeigte ihre räumliche Nähe zueinander. Bei der fluoreszenzspektroskopischen Untersuchung der an die katalytische Untereinheit A der V1ATPase gebundenen Fluoreszenzsonde N-[4-[7-(dimethylamino)-4-methyl]-cumarin-3-yl)]-maleimid (CM, Cumarinmaleimid) änderte sich die Sondenumgebung durch die Bindung von MgATP und MgADP bei einer Abnahme der Intensität um 20% (ATP) bzw. 25% (ADP) hin zum hydrophileren Milieu. Die Bindung von MgAMP-PNP und MgADP+Pi bewirkte keine Verschiebung des Fluoreszenzmaximums, die Intensität der Emission erhöhte sich um 10% (ADP+Pi) bzw. 25% (AMP-PNP). Die Untersuchung der intrinsischen Fluoreszenz des in den Untereinheiten A, B, C und H enthaltenen Tryptophans ergab in Abhängigkeit von der Bindung eines Nukleotids eine Abnahme der Fluoreszenzintensität um 50% in Anwesenheit von MgADP+Pi bzw. 65% (MgADP) und 72% (MgATP). In Anwesenheit des nicht hydrolysierbaren Nukleotidanalogons MgAMP-PNP war die Abnahme um 77% am stärksten. Dabei fand keine durch Verschiebung der Emissionsmaxima messbare Änderung der Umgebung der Tryptophane statt. Die Fluoreszenzspektroskopie verdeutlichte die Kopplung von der Nukleotidbindung am A3B3-"Köpfchen" der V1ATPase über die Stieluntereinheiten zum membrangebundenen VO-Teil bis zu dem daraus resultierenden Transport von Ionen über die Membran.
By detachment of single subunits of V1ATPase (A3:B3:C:D:E:F:G2:Hy) from Manduca Sexta with the detergent Lauryldimethyloxide (LDAO) hydrolytical active subcomplexes could be won. These were A3B3DEG, A3B3HEGF, A3B3EG and A3B3DEGH with a hydrolytic activity of 176 %, 53 %, 58% and 47% relative to the V1ATPase, respectively. The A3B3EG-subcomplex, which was minimum necessary for hydrolysis, was examined through electron microscopy in regard to the location of the stalkforming subunits E and G. Image analysis of the uranyl-acetate negative stained A3B3EG-molecule showed six hexagonal and alternating arranged subunits A and B. In the center of the hexamere subunits E and G were located. The A3B3EG-subcomplex was also compared in its behavior of limited tryptic digestion to the complete V1ATPase from M. sexta and subunit E from the yeast Saccharomyces cerevisiae. While the unprotected subunit E from S. cerevisiae was digested within few minutes, the partial protection by the catalytic subunits A and B delayed cleaving of the subunit E in the A3B3EG-subcomplex. Subunit E was shielded by the stalk-subunits C, D, F, G and H against proteolysis in the complete V1ATPase, so digestion was further prolongated. In order to identify the topology of the V1ATPase the zero-length crosslinker CuCl2 was used, resulting in the cross-linking formations A—B—E, A—B—D—E and D—E. Crosslinking with the photoaffinity label and nucleotide analogue P1,P4-Di-(3';-Biotinyl-8-azidoadenosine-5';-)-tetraphosphate (8-DiN3-DiB-AP4A) caused the products E—G, A—F(—G), A—B—E—G and D—E. The product of the central coupling element E and the stalk subunit D, which appeared with both crosslinkers, showed their proximity to each other. During the fluorescence-spectroscopic investigation of the fluorescence probe N-[4-[7-(dimethylamino)-4-methyl]-coumarine-3-yl)]-maleimide (CM, Coumarinmaleimide), which was bound to the catalytic subunit A of the V1ATPase, the probe neighborhood changed by the addition of MgATP and MgADP with a reduction of the intensity of approx. 20% (ATP) resp. 25% (ADP) to a more hydrophilic environment. Binding of MgAMP-PNP and MgADP+Pi did not cause any shift of the fluorescence maximum, the intensity of the emission increased by 10% (ADP+Pi) resp. 25% (AMP-PNP). The examination of the intrinsic fluorescence of the tryptophanes contained in the subunits A, B, C and H resulted in a reduction of the fluorescence intensity depending on the binding of a nucleotide at approx. 50% in presence of MgADP+Pi resp. 65% (MgADP) and 72% (MgATP). In presence of the not hydrolyzable nucleotide analogue MgAMP-PNP the decrease was highest with 77%. No changes of the environment of the tryptophanes measurable by shift of the emission maxima took place. Fluorescence spectroscopy showed the coupling of the nucleotide binding site at the A3B3-"headpiece" of the V1ATPase via the stalk subunits to the membrane-bound VO-part with the resulting transport of ions across the membrane.
