Abstract: Die biologisch wirksamen Acylphloroglucine Myrtucommulon A – I aus den Myrtengewächsen M. communis und C. scabrida wurden als potentielle Leitstrukturen zur Behandlung von Entzündungskrankheiten und Krebs identifiziert. In einer fünfstufigen Synthese gelang es, die Myrtucommulone und deren Strukturderivate mit Gesamtausbeuten bis zu 50 % darzustellen. Der symmetrische Aufbau der Naturstoffe erlaubte eine modulare Synthese mit einem hocheffizienten konvergenten Schlüßelschritt. Aus Phloroglucin als Grundbaustein wurden sowohl die Syncarpinäure-Einheit als auch der zentrale Acylphloroglucin-Kern unter minimalem Einsatz von Schutzgruppen und Verzicht auf Redoxreaktionen hergestellt und mittels einer basenkatalysierten doppelten Michael-Friedel-Crafts-Alkylierung verknüpft, um das vollständig funktionalisierte Kohlenstoffgerüst zu erzeugen. Zum Zwecke der Struktur-Wirkungs-Untersuchung wurde durch Variation der Acylgruppen, der Alkylgruppen und der 1,3-Diketon-Einheiten circa 30 verschieden Strukturderivate hergestellt und auf ihre Wirkung als duale Inhibitoren der mPGES-1 und der 5-LO getestet. Darüber hinaus wurden starke apoptoseinduzierende Wirkungen gegen mehrere Krebszellinien für einen großteil der Myrtucommulon-Strukturanaloga festgestellt. Die Wirkung einiger Strukturderivate konnte im Falle der Entzündungshemmung im Ver- gleich zur Leitstruktur Myrtucommulon A (IC50(mPGES) = 1 μmol/L) um ein Vielfaches verbessert werden.
The biologically active acylphloroglucinols myrtucommulone A – I from M. communis, and C. scabrida (Myrtaceae) were identified as lead compounds for the treatment of inflammation and cancer. The synthesis of the myrtucommulones and their analogues succeeded in five steps with overall yields up to 50 %. The symmetrical structure of the natural compounds allowed a modular synthesis with a highly efficient convergent key step. Starting from phloroglucinol as the basic module, the syncarpic acid units and the acylphloroglucinol core were synthesized with minimal use of protecting groups and without the use of redox reactions. These were then conected by a base catalyzed double Michael-Friedel-Crafts alkylation to generate the fully functionalized carbon framework. For the scope of structure-activity-investigations a set of ca. 30 structural analogues were synthesized by variation of the acyl, the alkyl an the 1,3-dicarbonyl units. The biological evaluation comprised the dual inhibition of mPGES-1 and 5-LO as well the apoptotic activity against severall cancer cell lines. The activity of some myrtucommulone analogues as inflammation inhibitors could be increased dramatically in comparison to the parent myrtucommulone A (IC50(mPGES) = 1 μmol/L).
