Abstract: Zur Gewinnung neuer Erkenntnisse über die Regulation des Stoffwechsels von Bacillus subtilis wurde die Rolle des globalen Regulatorproteins CcpA untersucht. Dies erfolgte einerseits anhand metabolischer Charakterisierung und vergleichender Analyse der Expressionsmuster kataboler Gene von B. subtilis 168 bei Nutzung unterschiedlicher Substrate, sowie andererseits anhand der metabolischen Charakterisierung verschiedener ccpA-Mutanten. Es wurde gezeigt, dass die Aktivität der beiden Stoffwechselwege Glykolyse und Pentosephosphatweg nicht auf Transkriptebene gesteuert wird, sondern offenbar auf metabolischer Ebene durch die Verfügbarkeit intrazellulärer Metabolite beeinflusst wird. Der Überflussmetabolismus von Bacillus subtilis wird offenbar sowohl auf Transkript- als auch auf Metabolom-Ebene reguliert, wobei die Ausschaltung der Katabolitenrepression die Bildung von Überflussmetaboliten nicht vollständig unterdrückt. Die metabolische Aktivität des Tricarbonsäurezyklus ist dagegen hauptsächlich auf Transkriptebene über die synergistische Repression der Citrat-Synthase und Aconitase durch CcpA und den Repressor CcpC reguliert. Die Rolle von CcpA als globalem Regulator des Zentralmetabolismus in Abhängigkeit des Nährstoffangebots wurde in dieser Arbeit zwar bestätigt, jedoch besitzt dieser Effektor offenbar eine eher unterstützende Funktion bei der Regulation des Stoffwechsels und übt keine vollständige Kontrolle über einzelne Stoffwechselwege aus.
The role of the global regulator protein CcpA was investigated to obtain deeper insight into the regulation of Bacillus subtilis metabolism. This was achieved on the one hand by metabolic characterization and parallel analysis of gene expression patterns in B. subtilis 168 growing on different carbon sources and on the other hand by metabolic characterization of ccpA mutants. The activity of glycolysis and pentose phosphate pathway was shown to be regulated independently from the transcriptional level and glucose induced catabolite repression but was affected by the availability of metabolic intermediates and therefore is presumed to be mainly controlled on the metabolic level. The overflow metabolism pathways of Bacillus subtilis are obviously regulated on both, transcriptional and metabolic level, while elimination of carbon catabolite repression did not completely suppress the formation of overflow metabolites. The metabolic activity of the Krebs-cycle was shown to be mainly regulated on the transcriptional level by synergistic repression of citrate synthase and aconitase by CcpA and the repressor Protein CcpC. On the one hand the role of CcpA as a global regulator was confirmed by these studies but on the other hand the observations showed a more supporting role of CcpA in the regulation of the Bacillus subtilis central metabolism, precisely because this regulator did not exhibit total control on the metabolic activity of any pathway.
